摘要
目的研究极性有机溶剂辅助阴、阳离子交换色谱串接组合纯化蛋白质的可行性。方法以溶栓素为纯化对象 ,采用极性有机溶剂辅助阴、阳离子交换色谱串接组合与常规方法的纯化结果相对比。结果聚丙烯酰胺凝胶电泳考马斯亮蓝染色法结果显示 ,以含低浓度乙醇的缓冲液进行相同的离子交换色谱 ,具有较好的分离效果。结论极性有机溶剂辅助阴、阳离子交换色谱有效串接组合是一种较好的组合纯化方式 ,但是这种方法使用的前提条件 。
PurposeTo explore the possibility of combination of anion and cation exchange chromatography by using the polarity organic solvents in eluent.MethodsPurification results were compared between the combination chromatography of anion and cation exchange assisted with polarity organic solvents and the routine combination chromatography.ResultsIt was shown by polyacryl amide gel electrophoresis that the combination chromatography of anion and cation exchange assisted with ethanol had better purifying ability than that of the routine combination chromatography.ConclusionCombination chromatography of anion and cation exchange assisted with polarity organic solvents could obtain good results in the purification of proteins if the organic solvents had no effect on the solubility and activity under the chromatography conditions.
