摘要 目的探讨RNA干扰Th17关键转录因子ROR-γt表达后,对大鼠肾移植慢性排斥模型中Th17细胞功能以及慢性排斥反应的影响。方法构建ROR-γt特异性干扰腺病毒,将其通过尾静脉注入大鼠慢性排斥反应模型,监测大鼠血肌酐变化,酶联免疫吸附试验检测Th17相关细胞因子IL-17、IL-6和TNF-γ改变。4周后检测大鼠肾组织病理改变。结果成功构建ROR-γt特异性干扰腺病毒pSES-HUS-ROR-γt。大鼠慢性排斥反应模型尾静脉注入pSES-HUS-ROR-γt,大鼠血清肌酐水平下降,IL-17、IL-6和TNF-γ水平下降,肾小球纤维化病变减轻。结论 pSES-HUS-ROR-γt特异性干扰腺病毒可以抑制Th17细胞功能,减轻肾移植慢性排斥反应。 This study was performed to explore the changes of Th17 function and kidney transplantationchronic rejection reaction in rat model after transfection with p SES-HUS-ROR-γt interference adenovirus.ROR-γt interference adenovirus was constructed, and injected into kidney transplantation chronic rejection reactionrat model by tail vein. The change of Cr(serum creatinine) was monitored; Th17 related cytokines IL-17, IL-6 andTNF-γ(tumor necrosis factor-γ) were detected by enzyme-linked immunosorbent assay(ELISA); kidneypathological change was analyzed 4 weeks after the injection. Data showed that ROR-γt interference adenovirus wasconstructed and injected into kidney transplantation chronic rejection reaction rat model successfully. Inexperimental group, the levels of serum Cr, IL-17, IL-6 and TNF-γ were decreased, and glomerular fibrosis lesionsalleviated. In conclusion, p SES-HUS-ROR-γt interference adenovirus can inhibit the function of Th17 cells, andcan reduce the kidney transplantation chronic rejection reaction in rat model.
