摘要
用复合酶法提取海带中的褐藻酸钠,并对工艺进行优化。通过对单因素实验和正交试验结果的分析,确定褐藻酸钠最佳提取工艺为:粉碎的海带经过浸泡过滤后,加入海带干重1%(m/m)的纤维素酶和1%(m/m)的果胶酶,55℃(p H值4.5)水解2h,再加入1%(m/m)的木瓜蛋白酶,85℃(p H值8.5)水解2 h。此工艺的提取率高,产品纯度高,是对海带多糖传统提纯工艺的革新。
The method of compound enzymes was used to extract sodium alginate from brown seaweeds,with its process optimized. The analysis of the results of single-factor and orthogonal tests indicated that the optimum conditions of sodium alginate extraction were obtained as follows: after soaking and filtering of the dusty brown seaweeds,1%( m / m) cellulase and1%( m / m) pectinase( at 55℃ and pH 4. 5) were added to extract for 2 h,followed by 1%( m / m) protease( at 85℃ and pH 8. 5) for 2h. The compound enzymatic extraction has advantages of high extraction ratio and fine purity quotient,which becomes an innovation of the traditional extraction- purification processes of polysaccharide in the brown seaweeds.
