柴胡皂苷d对肝星状细胞ERα和ERβ mRNA水平的调节 被引量:14

Regulation of Saikosaponin-d on levels of ERα,ERβ mRNA in rat hepatic stellate cells

摘要 目的:研究柴胡皂苷d对大鼠肝星状细胞ERα、ERβmRNA的调节作用,探讨其对肝纤维化的作用机制。方法:体外培养大鼠肝星状细胞系HSC-T6,采用实时荧光定量RT-PCR方法,观察柴胡皂苷d对两种雌激素受体亚型表达的影响。采用MTT法,观察雌二醇、柴胡皂苷d对HSC-T6细胞增殖的影响,并为后续实验确定合适的实验用药物剂量和用药时间。结果:柴胡皂苷d能明显提高HSC-T6细胞ERα、βmRNA表达,且呈剂量与时间依赖性,但作用较雌二醇弱,这种作用可被ER完全拮抗剂ICI-182.780抑制。通过对半数抑制率的计算,得出柴胡皂苷d和雌二醇半数抑制率(IC50)的浓度分别为5μmol/l和1μmol/l。结论:推测柴胡皂苷d有雌激素受体调节剂的作用,可能是一种雌激素调节剂。 Objective: To study the effect of saikosaponin-d on rat hepatic stellate cell of ER α,ER β mRNA regulation,of phytoestrogens on hepatic fibrosis,clinical application and provide theoretical basis for plant estrogen. Method: In vitro culture of rat hepatic stellate cell line HSC-T6,using real-time fluorescence quantitative RT-PCR method,saikosaponin-d was two estrogen receptor subtypes expression influence.Determined by MTT method,observation of saikosaponin-dand estradiol affect HSC-T6 cell proliferation,and for follow-up experiments to determine the appropriate experimental drug dosage and duration. Results: Saikosaponin-d could increase the expression of α,β mRNA ER HSC-T6 cells,and a dose and time dependent,but compared to estradiol,the role of the weak,this effect can be fully ER antagonist ICI-182. 780. Through calculation of half the inhibition rate of saikosaponin-d and estradiol inhibition( IC50) concentrations respectively 5μmol/l and 1μmol / l. Conclusion: Suppose that saikosaponin-d has the effect of estrogen receptor modulators,may be an estrogen regulator.
