摘要
目的:观察白藜芦醇对大鼠酒精性脂肪肝的作用并探讨相关的作用机制。方法:用白酒灌胃诱导建立大鼠酒精性脂肪肝(AFL)模型,分设空白对照组、模型组、硫普罗宁(阳性对照)组、白藜芦醇(62.5、125、250mg/kg)治疗组。白藜芦醇干预9周后,称取大鼠体质量、体脂和肝脏重量计算肝指数与体脂比,分离血清测定血脂、转氨酶、NO和肝组织匀浆测定SOD、MDA指标,病理切片观察肝脏的病理变化。结果:与模型组相比,白藜芦醇(125、250mg/kg)治疗组大鼠的体脂量比、肝指数、丙氨酸转氨酶(ALT)、甘油三脂(TG)、丙二醛(MDA)的含量显著降低,超氧化物歧化酶(SOD)活性显著增加;白藜芦醇(62.5、125、250mg/kg)治疗组天冬氨酸转氨酶(AST)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-D)、一氧化氮(NO)的含量显著降低,低密度脂蛋白胆固醇(HDL-D)含量显著增加,大鼠肝组织脂肪变明显减轻。结论:白藜芦醇具有降低转氨酶、血脂和抗氧化作用,减少肝细胞中的脂质沉积与肝损伤,改善白酒诱导的酒精性脂肪肝。
Objective: To observe the effect of resveratrol on alcoholic fatty liver( AFL) in rats and to explore the mechanism of related. Methods:Alcoholic fatty liver model was established by induced with liquor gavage,and rats were divided into resveratrol treatment groups including low-,medium-and high-dose group( 62. 5,125,250 mg / kg),corresponding control groups( blank group,model group and tiopronin positive control). After nine weeks of intervention by resveratrol,rats body weight,body fat and liver weight was used to weight to calculate the liver index and body fat ratio,the changes of serum lipid,transaminase indicator and NO in the serum were determined,SOD and MDA index of liver homogenate were measured,and the optical microscope was used to observe the degree of hepatiesteatosis. Results: Compared with model groups,the degree of rat liver index,ratio of body fat,ALT,TG and MDA were reduced remarkably,meanwhile the content of HDL-D and activity of SOD were enhanced obviously in the resveratrol( 125,250 mg / kg) treatment groups; the levels of AST,TC,LDL-Dand NO were decreased significantly,hepatiesteatosis in rats was reduced clearly in the resveratrol( 62. 5,125,250 mg / kg) treatment groups. Conclusion: Resveratrol has decrease transaminase,blood lipid and antioxidation,reduce lipid deposition in liver cells,and to improve the alcoholic fatty liver by alcohol induced.
