摘要
目的观察乳腺癌患者外周血及组织中白细胞介素(IL)-22及IL-22结合蛋白(IL-22BP)表达水平对乳腺癌发生发展的影响。方法采用酶联免疫吸附试验法(ELISA)检测95例乳腺癌患者和20例乳腺良性肿瘤患者血清IL-22和IL-22BP表达水平,用免疫组织化学(IHC)方法检测两者存乳腺癌组织和癌旁组织表达。结果乳腺癌患者外周血IL-22为(64.5±22.1)pg/ml,显著高于乳腺良性肿瘤患者外周血IL-22水平[(23.7±11.4)pg/ml,P〈0.01];乳腺癌患者外周血IL-22BP为(260.6±70.9)pg/ml,显著低于乳腺良性肿瘤患者外周血IL-22BP水平[(472.1±156.3)pg/ml,P〈0.01]。IL-22在乳腺癌组织阳性表达率为71.57%,在癌旁组织阳性表达率为24.21%,两者差异有统计学意义(x^2=42.71,P〈0.01)。IL-22BP在乳腺癌组织中的阳性表达率为27.37%,在癌旁组织中的阳性表达率为74.74%,两者差异有统计学意义(x^2=42.65,P〈0.01)。11.-22在细胞组织学Ⅲ级、有淋巴结转移、肿瘤最大径〉2cm、临床分期Ⅲ期、人类表皮生长因子受体-2(Her-2)过表达和细胞核增殖抗原(Ki-67)阳性率≥14%组明显升高(P〈0.01或P〈0.05)。而IL-22BP在上述组中明显降低(P〈0.01或P〈0.05)。结论IL-22的高表达或其拮抗蛋白IL-22BP的低表达,与乳腺癌的发生、发展及转移关系密切。
Objective To investigate the expression of interleukin (IL) -22 and IL -22 binding protein ( IL - 22BP) in patients with breast tumor and to analyze the relationship between them and patho- logical characteristics. Methods Ninety -five patients with breast cancer, and 20 patients with breast be- nign tumor were included. The concentrations of IL- 22 and IL -22BP in serum were measured by enzyme - linked immunosorbent assay (ELISA). Their expression in breast cancer tissues and non - can- cerous adjacent breast tissues were examined by immunohistochemistry. Statistical analysis of the relevance of IL - 22, IL - 22BP and clinicopathological variables was performed. Results The level of IL - 22 in pe- ripheral blood of patients with breast cancer was (64. 5 ± 22. 1 ) pg/ml, significantly higher than that in be- nign tumor [ (23.7±11.4) pg/ml] (P 〈 0. 01 ). The level of IL- 22BP in peripheral blood of patients with breast cancer was (260. 6 ± 70. 9 ) pg/ml, significantly lower than that in benign tumor [ (472. 1 ± 156. 3) pg/ml] (P 〈 0. 01 ). The expression rate of IL -22 was 71.57% in breast cancer tissues and 24.21% in non- cancerous adjacent breast tissues respectively (P〈 0. 01 ). The expression rate of IL-22BP was 27.37% in breast cancer tissues, and 74. 74% in non -cancerous adjacent breast tissues ( P 〈 0. 01 ). The expression of IL -22 and IL -22BP was significantly positively correlated with histologi- cal grade, axillary lymph nodes metastasis, larger tumor size, tumor stage, proliferation cell nuclear anti- gen (Ki-67) ≥ 14% and human epidermal growth factor rceeptor- 2 (Her- 2) positive status (P 〈 0. 01 or P 〈0. 05) , but not with age at diagnosis, menopausal status, estrogen receptor (ER) status and progesterone receptor (PR) status. Conclusion The high expression of IL -22 and low expression of 11,- 22BP in patients with breast cancer might be closely related with the pathogenesis and development of breast cancer.
