Expression of N-myc downstream-regulated gene 1 in renal cell carcinoma and its clinical significance

摘要 目的 探讨N-myc下游调节基因1(NDRG1)在肾细胞癌(RCC)组织中的表达及其与RCC临床病理学特征的相关性.方法 采用免疫组织化学PV-6000法检测128例手术切除RCC组织及20例癌旁肾组织中NDRG1蛋白的表达,分析NDRG1阳性表达与肾癌病理类型、组织学分级和临床分期等参数之间的关系.结果 NDRG1在正常肾组织中和RCC组织的阳性表达率分别为100%(20/20)和54.7% (70/128),组间差异有统计学意义(P<0.05).随着RCC临床分期、组织学分级的增加,NDRG1阳性表达程度逐渐减低,各组间差异均有统计学意义(P<0.05),NDRG1在淋巴结转移组阳性表达率显著低于无转移组(P<0.05).结论 NDRG1蛋白在RCC的表达程度降低且与其分期、分级等病理参数关系密切,提示NDRG1在RCC发生、发展中可能发挥抑癌基因的作用. Objective To detect the expression of N-myc downstream-regulated gene 1 (NDRG1) in renal cell carcinoma (RCC) and to investigate the relation between NDRG1 protein and clinicalopathological features of RCC.Methods The immunohistochemical PV-6000 method was used to detect the expressed level of NDRG1 protein in 128 cases of RCC and 20 cases of normal renal tissues.The correlation between the NDRG1 expression and clinicopathological parameters was analyzed.Results The positive expression rate of NDRG1 protein in RCC (54.7%) was decreased significantly as compared with that in normal renal tissues (100%) (P 〈 0.05).With the increase of clinical stage and histological grade of RCC,the positive expression rate of NDRG1 was significantly decreased (P 〈 0.05).Conclusion NDRG1 may play an anti-oncogene role in the carcinogenesis and progression of RCC with a low expression level in RCC and a close relation between NDRG1 and clinicalopathological parameters of RCC.
