卵黄抗体检测评估SPF鸡对鸡传染性贫血病病毒的感染状态研究 被引量:2

摘要 鸡传染性贫血病病毒(CIAV)是禽弱毒疫苗中常见外源病毒之一,选择未被CIAV污染的SPF鸡胚是生产合格疫苗的关键环节,疫苗生产企业通常通过检测SPF鸡血清中CIAV的抗体来评估CIAV感染状况,本研究以取样更为方便的卵黄抗体检测来评估SPF鸡群中CIAV感染状态。将40只SPF鸡于19周龄时人工接种CIAV,同时30只SPF鸡饲养于另一隔离环境内作为空白对照。23周龄时每只鸡分别采集种蛋和血清并一一对应,然后将卵黄分别进行1∶10、1∶20、1∶30稀释后与血清同时检测CIAV抗体,结果发现卵黄抗体1∶20倍稀释后判定结果与血清抗体吻合率最高。对70只SPF鸡从25~34周龄进行连续10周观察,结果显示卵黄抗体检测结果与血清抗体检测结果吻合率达91.57%。建议疫苗生产企业可通过抽检卵黄抗体来判断SPF鸡群的CIAV洁净度。 Chicken infectious anemia virus(CIAV)is the most frequent exogenous virus that contaminates attenuated vaccines. It is extremely important to select CIAV-free SPF chicken embryos. Generally evaluation of CIAV infection is used for detecting CIAV antibody in SPF chickens. This study attempted to evaluate CIAV infection by replacing serum antibody detection with yolk antibody detection. Forty 19- week- age SPF chickens were artificially inoculated with CIAV,with 30 SPF chickens raised in another isolation environment as a blank control group. Eggs and serums for each 23-week-age chickens were sampled to establish one-to-one correspondence,then the yolks were diluted separately to the ratios of 1∶10,1∶20,1∶40 and were detected together with serums. The results showed that the evaluation result based on yolk antibody detection at the dilution rate of 1∶20 had the highest coincidence rate with that based on serum antibody detection. At the dilution ratio of 1∶20 for yolk antibody,70 chickens were continuously observed for 10 weeks from 25 to 34 weeks of age. The results demonstrated that based on the serum antibody detection or the yolk antibody detection,the coincidence rate was 91.57%. Accordingly,it was suggested that vaccine producer could estimate CIAV cleanliness in poultry farm by detecting egg yolk antibody.
