复方三七消痛软膏治疗兔膝骨性关节炎的疗效及其对软骨细胞caspase-3表达的影响

Sanqi Xiaotong Compound Cream on expression of apoptotic cells and caspase-3 of rabbits with knee osteoarthritis

目的观察外擦复方三七消痛软膏治疗兔膝骨性关节炎疗效及其对软骨细胞caspase-3表达的影响。方法 15只新西兰大白兔随机分为A、B、C组(每组各5只);A、B 2组均行右侧膝关节ACLT术,C组不予任何处理;术后6周A组以复方三七消痛软膏外擦并结合拍打患处,B、C2组不予任何干预,正常饲养。干预2周后取材,HE染色法观察软骨组织形态并行改良Mankin评分,免疫组化法检测软骨细胞caspase-3表达。结果 1)改良Mankin评分比较:A组(5.15±1.14)明显低于B组(8.50±1.10),但明显高于C组(2.40±1.35),差异均有统计学意义(P<0.01);2)caspase-3 IOD:与B组(23 865.80±7 666.48)相比,A组(2 994.35±1 215.28)阳性表达明显下降(P<0.01);与C组相比(453.80±265.24),B组阳性表达显著升高(P<0.01)。结论外擦复方三七消痛软膏结合拍打兔膝骨性关节炎模型可以延缓兔膝骨性关节炎的进展,其机制可能与调节软骨细胞caspase-3的表达,抑制软骨细胞凋亡有关。

Objective To observe the treatment effects of wiping sanqixiaotong compound creamon the expression of apoptosis cells and caspase-3 in rabbits with knee osteoarthritis. Methods The 15 New Zealand rabbits with knee osteoarthritis were randomly divided into A group（ n = 5） and B group（ n = 5）,and C group（ n = 5）. The former 2groups were modeled as anterior cruciate ligament transaction（ ACLT）. Rabbits of the A group were received wiping Sanqi Xiaotong compound cream on knee joint 6 weeks afeter operation and continued for 2 weeks,while those in the B group and C group were administered with not any intervention for 2 weeks. They were tested with the modified Markin,observed with hematoxylin and eosin staining. Immunohistochemistry was used to investigate the expression of caspase-3 in articular cartilage. Results The modified Markin score from the A group（ 5. 15 ± 1. 14） lower than the B group（ 8. 50 ± 1. 10）,but significantly higher than group C（ 2. 40 ± 1. 35）,the difference had statistical significance（ P〈0. 01）. caspase-3 IOD： The expression of caspase-3 decreased in the A group（ 2 994. 35 ± 1 215. 28） compared with the B group（ 23 865. 80 ± 7 666. 48）（ P〈0. 01）. The expresion of caspase-3 inceased in the B group compared with C group（ 453. 80 ± 265. 24）（ P〈0. 01）. Conclusion Sanqi Xiaotong compound cream on rabbits could delay the progress of knee osteoarthritis,which may regulate the expression of apoptotic cells and caspase-3.
