C-myc基因调控COPD大鼠气道重塑及苗药对其干预的研究

Study on the effects of Miao Nationality's Herbs And C-myc on airway redmeling of COPD rats

目的观察慢性阻塞性肺疾病(COPD)大鼠气道重塑模型中原癌(C-myc)基因mRNA及蛋白的表达情况,探讨C-myc基因与气道重塑的关系及苗药对COPD大鼠气道重塑的干预。方法 90只雄性wistar大鼠随机分为正常组、COPD模型组、苗药组方干预组各30只。COPD大鼠气道重塑的模型制作采用熏烟法,并每月2次在气管内注射脂多糖的方法建立;苗药组方干预组大鼠于造模成功后给予灌胃治疗。处死大鼠后,分别运用实时荧光定量法﹙RT-qPCR﹚、免疫组化法检测各组大鼠气道及肺组织中原癌基因mRNA及蛋白的表达情况。结果与正常组相比较,COPD模型组与苗药组方干预组气道及肺组织中原癌基因mRNA含量及蛋白阳性表达率明显增高﹙P<0.05﹚;与COPD模型组相比,苗药组方干预组原癌基因mRNA及蛋白阳性表达率明显降低(P<0.05)。结论 COPD大鼠气道重塑这一病理变化可能受C-myc基因的调控;苗药对COPD大鼠气道重塑模型的建立有一定程度的干预,苗药组方的应用可在一定程度上抑制C-myc基因和蛋白的表达。

Objective To observe the level changes of C-myc mRNA and protein in the rats＇ lung tissue,and to explore the relationship between C-myc and airway remodeling,and explaining the effect of Miao nationality＇s herb on the airway remodeling.Methods 90 wistar male rats were randomly divided into NS group,COPD group,Miao nationality′s herb treatment group,and 30 in each group.The COPD rat model was built with compound factors：cigarette smoke plus intratracheal instillation of lipopolysaccharide（LPS）.After duplicating the mold,the rats in Miao nationality′s herb treatment group were treated by stomach.All animals were put to death and samples were collected.The expression of C-myc mRNA and protein content in the rats＇ lung tissue were detected in both groups by immunohistochemistry and RT-qPCR.Results Compared with NS group,the expression of C-myc mRNA and protein content in COPD group and Miao nationality＇s herb treatment group showed a increasing trend,the difference was statistically significant（P〈0.05）.Compared with COPD group,the expression of C-myc mRNA and protein content in Miao nationality＇s herb treatment group showed a decreasing trend（P〈0.05）.Conclusion C-myc may play a role in airway remodeling in COPD rats.Miao nationality′s herb was effective in both reducing airway remodeling and decreasing the expression of C-myc mRNA and protein content.
