摘要
目的探讨上皮间质转化标志物锌指E-盒结合同源异形盒-1(ZEB1)、E-钙黏蛋白(E-cadherin)、N-钙黏蛋白(N-cadherin)在舌鳞癌组织中的表达及其临床意义。方法采用免疫组织化学Streptavidin-Peroxidase(SP)法检测68例舌鳞癌组织、63例舌癌癌旁组织和68例正常舌上皮组织中ZEB1、E-cadherin、和N-cadherin的表达情况,并分析其表达与患者临床病理特征及转移的关系。结果 ZEB1、E-cadherin蛋白在舌鳞癌组织中的阳性表达率低于癌旁组织、正常上皮组织(P<0.05);而N-cadherin蛋白在舌鳞癌组织中的阳性表达率高于癌旁组织、正常上皮组织(P<0.05)。舌鳞癌组织中ZEB1、E-cadherin和N-cadherin蛋白的阳性表达率在不同肿瘤TNM分期、肿瘤分化程度及有无淋巴结转移间比较差异有统计学意义(P<0.05)。舌癌组织中ZEB1蛋白的阳性表达与E-cadherin蛋白阳性表达呈正相关;ZEB1蛋白的阳性表达与N-cadherin阳性蛋白表达呈负相关。结论 ZEB1、E-cadherin和N-cadherin在舌鳞癌演变过程起重要的作用,可能参与了舌鳞癌侵袭和转移的过程。
Objective To investigate the expressions of Epithelial mesenchymal transition(EMT)markers Zinc Finger E-Box Binding Homeobox 1(ZEB1),E-cadherin and N-cadherin in tongue squamous cell carcinoma,and their clinical significances thereof.Methods Immunohistochemical streptavidin-peroxidase method was applied to detect expressions of ZEB1,E-cadherin and N-cadherin in 68 samples of tongue squamous cell carcinoma,63 samples of tongue cancer adjacent tissues and 68 samples of normal tongue epithelium tissues.Correlations of expressions of ZEB1,E-cadherin and N-cadherin to clinical pathological parameters and metastasis were analyzed in three groups.Results The expression positive rates of ZEB1 and E-cadherin were significantly lower in tongue squamous cell carcinoma than those in tongue cancer adjacent tissues and normal tongue epithelium tissues(P〈0.05).The expression positive rates of N-cadherin were significantly higher in tongue squamous cell carcinoma than those in tongue cancer adjacent tissues and normal tongue epithelium tissues(P〈0.05).There were statistical significantly differences in the expression of ZEB1,E-cadherin and N-cadherin between different TNM stages,differentiation and lymph node metastasis in tongue squamous cell carcinoma(P〈0.05).The positive expression of ZEB1 was positively correlated with the positive expression of E-cadherin.The positive expression of ZEB1 was negatively correlated with the positive expression of E-cadherin.Conclusion ZEB1,E-cadherin and N-cadherin play an important role in evolution of tongue squamous cell,it may be involved in the process of tongue squamous cell invasion and metastasis.
