摘要
种子休眠性是花生重要的农艺性状。为了挖掘控制花生种子休眠的重要基因,以花育52号为试材,通过转录组测序获得生长素抑制蛋白基因的全长,该基因全长为918bp,开放阅读框为363bp,编码121个氨基酸,推导出的蛋白质分子量为13.44 k Da,理论等电点p I=9.64。序列比对表明,该序列与花生生长素抑制蛋白基因Ah ARP一致,确认该c DNA序列为编码花生生长素抑制蛋白基因的全长c DNA序列。荧光定量PCR结果表明,Ah ARP基因在种子休眠阶段表达量较高,种子休眠解除后表达量降低;乙烯利释放吸涨种子休眠过程中,Ah ARP基因受到明显诱导;说明Ah ARP基因可能与花生种子休眠有关。Ah ARP基因的序列、结构、性质以及功能的初步分析,为进一步研究该基因在花生种子休眠中的功能机制奠定了基础。
Peanut( Arachis hypogaea L.) is an important oil and economic crop in China.Peanut Seed dormancy is an important agronomic trait which can affect quality,yield and seed quality in agricultural production.To dig important functional genes controlling seed dormancy,full-length of auxin-reprssed protein gene was screened from seed of peanut cultivar Huayu52 by RNA-seq,and designated as Ah ARP in this article.The whole sequence of Ah ARP was 918 bp and its open reading frame was 363 bp,encoding a 121 amino acid residues.The deduced protein molecular weight was13.44 KD and its theoretical isoelectric point was 9.64.Sequenc alignment of the deduced amino acids of Ah ARP revealed high similarity with auxin-repressed protein gene( AAZ20292.1) of peanut.The results of Real-time RT-PCR showed that Ah ARP gene expressed at a higher level in dormant variety than in non-dormant variety; The expression of Ah ARP in dormancy-released seed was induced distinctly by ethephon.That indicated Ah ARP may be correlated to the seed dormancy of peanut.According to the gene sequence and preliminarily predicting structure,nature and functions of AhRP gene.The study have acquired valuable information which can be used to study the biological function of the Ah ARP gene related to peanut seed dormancy.
