自拟益气养阴清热利湿方药对IgA肾病大鼠肾脏局部激肽释放酶-激肽系统的影响 被引量:5

摘要 目的:探讨益气养阴清热利湿方药对IgA肾病大鼠的治疗作用及对肾脏局部激肽释放酶-激肽系统系统的影响。方法:将SD大鼠随机分为4组,对照组、模型组、西药组、中药组,各组按相应药物剂量灌胃;造模前和第7、9、13、17周末检测24h尿蛋白定量,第17周末行肾组织IgA免疫荧光检测,并应用RT-PCR法检测肾组织KLK1和BK的表达水平。结果:与模型组比较,中药组、西药组IgA免疫荧光强度显著下降,KLK1和BK的表达水平显著增高(P<0.05)。结论:益气养阴清热利湿方药可能是通过激活肾脏局部KKS,达到延缓肾脏病理进展,发挥对IgA肾病的治疗作用。 Objective: To investigate the effect of Supplementing Qi and Nourishing Yin-Clearing Heat and Draining Dampness Formula on the expression of the renal local KKS. Methods: SD rats were randomly divided into 4 groups: control group,model group,Western medicine group and Chinese medicine group,which afforded with the medicine respectively. Before replication of the model,at end of the seventh,ninth,thirteenth and seventeenth week,the levels of 24-hour urinary protein were tested. At the end of the seventeenth week,Ig A in the kidney tissue was detected by immunofluorescence and kallikrein 1( KLK1) and bradykinin( BK) detected with RT-PCR. Results: In Western medicine and Chinese medicine groups,the Ig A immunofluorescence was significantly decreased in contrast with model groups. Compared with model group,the levels of the KLK1 and BK were significantly increased( P〈0. 05). Conclusion: Supplementing Qi and Nourishing Yin-Clearing Heat and Draining Dampness Formula could treat Ig A nephropathy rats effectively by the activation of renal local KKS.
