基于FXR探讨茵栀黄注射液利胆退黄的机制研究 被引量:18

摘要 目的:通过研究茵栀黄注射液对肝内胆汁淤积湿热证大鼠FXR、BSEP、NTCP蛋白和基因表达的影响,探讨茵栀黄注射液的作用靶点及机制。方法:51只雄性SD大鼠随机分为正常对照组(7只)、模型组(7只)、茵栀黄注射液组(7只)、GW4064组(7只)、Guggulsterone组(7只)、茵栀黄+GW4064组(8只)、茵栀黄+Guggulsterone组(8只)共7组。造模干预后观察各组大鼠肝组织FXR、BSEP、NTCP蛋白和基因的表达。结果:茵栀黄注射液可以上调FXR蛋白和基因的表达,P<0.05。FXR和BSEP在蛋白和基因表达量上存在显著相关,P<0.05。结论:茵栀黄注射液通过干预FXR上调BSEP利胆退黄治疗肝内胆汁淤积。 Objective: To study the effect of Yinzhihuang Injection on FXR,BSEP and NTCP expressions to understand the specific targets in the treatment of intrahepatic cholestasis. Methods: A total of 51 male SD rats were randomly divided into normal group( 7),model group( 7),Yinzhihuang group( 7),GW4064 group( 7),Guggulsterone group( 7),Yinzhihuang + GW4064 group( 8),Yinzhihuang + Guggulsterone group( 8). After establishing the disease and syndrome combined model after drug intervention,we observed the effect of Yinzhihuang Injection on liver's FXR,BSEP and NTCP expressions. Results: Yinzhihuang Injection can increase FXR protein and gene expression,P 0. 05. FXR and BSEP have a significant correlation on the protein and gene expression. Conclusion: Yinzhihuang Injection can increase the expression of FXR to raise BSEP in the treatment of intrahepatic cholestasis.
