摘要
目的:探讨不同缺氧时间点对小鼠心脏成纤维细胞旁分泌TGF-β1、BMP-2蛋白和mRNA的影响。方法:将体外分离培养的第2代小鼠心脏成纤维细胞,分别缺氧0、12、24、48 h。ELISA法和RT-PCR法检测不同时间点心脏成纤维细胞旁分泌TGF-β1、BMP-2的蛋白和mRNA水平。结果:相较于缺氧0 h,缺氧12 h后TGF-β1的蛋白和mRNA表达呈上调趋势,但24 h、48 h后逐步下调;而BMP-2在缺氧12 h、24 h后,蛋白和mRNA表达均逐步上调,48 h后mRNA水平下调。与缺氧24 h相比,缺氧48 h后TGF-β1和BMP-2的蛋白以及mRNA表达均下调。结论:不同缺氧时间诱导的心脏成纤维细胞旁分泌TGF-β1和BMP-2的变化,可能参与了心肌纤维化的病理过程。
Objective: To investigate the paracrine of TGF-β1 and BMP-2 in mice cardiac fibroblasts at different hypoxia time points in vitro. Methods: The mice cardiac fibroblasts were isolated and cultured in vitro. The second-passage cells were harvested and induced hypoxia for 0 h,12 h,24 h and 48 h. ELISA and RT-PCR were used to evaluated the levels of TGF-β1 and BMP-2 respectively. Results: Compared with hypoxia for 0 h,the protein and mRNA expression of TGF-β1 in hypoxia for 12 h had an increasing trend,but from 24 h on,the results tuned gradually lower. Meanwhile,the BMP-2 in both protein and mRNA level were up-regulated in hypoxia for 12 h and 24 h,however 48 h later,the results decreased. Compared with hypoxia for 24 h,the protein and mRNA expressions of TGF-β1 and BMP-2 in hypoxia for 48 h were reduced. Conclusion: The expressions of TGF-β1 and BMP-2 in cardiac fibroblasts were exchanged under hypoxia,which might involve in the pathological process of myocardial fibrosis.
