失荅剌知丸对脑缺血再灌注大鼠脑梗死体积及缺血侧脑组织NF-κB表达的影响 被引量:4

摘要 目的:观察失荅剌知丸对脑缺血再灌注大鼠缺血侧脑组织NF-κB表达的影响。方法:将96只SD雄性大鼠随机分为假手术组、缺血再灌注组、金纳多阳性药物对照组、失答剌知丸低、中、高剂量组,后5组采用线栓法制备大脑中动脉闭塞(MCAO)模型,缺血2 h后再灌注,术后分别于12 h、24 h、72 h取材,共16组,每组6只。TTC染色法检测脑组织梗死体积;干湿重法测脑含水量;逆转录-聚合酶链反应技术(RT-PCR)分析测定NF-κBmRNA的活性表达。结果:与假手术组比较,缺血再灌注组及各给药组各时间点大鼠脑梗死体积、脑组织含水量及NF-κBmRNA的表达均显著增加(P<0.01);与IR组比较,各给药组各时间点脑梗死体积、脑组织含水量及NF-κBmRNA的表达均显著降低(P<0.01);与GC组比较,失答剌知丸各时间点脑梗死体积、脑组织含水量及NF-κBmRNA的表达有统计学意义(P<0.05或P<0.01),其中SH组各时间点脑梗死体积、脑组织含水量及NF-κBmRNA的表达显著减少(P<0.01)。结论:失荅剌知丸不仅能够减少大鼠脑梗死体积及脑组织含水量,还能够明显降低脑组织NF-κB的表达,抑制炎症反应,其发挥神经保护的作用机制可能与抑制炎症反应有关。 Objective: To observe the impact of Shida Lazhi Pill on the expression of NF-κB in cerebral ischemia reperfusion model. Methods: A total of 96 SD male rats were used in this study. They were randomly divided into six groups,including control group,ischemia-reperfusion group,ginaton group and the low,medium,high dose groups of Shida Lazhi Pill. The suture method was used to prepare MCAO model in last five groups. Make ischemia reperfusion after 2 h in MCAO model and take the tissue samples respectively in the 12 h,24 h,72 h after operation,a total of 16 groups,each group six. TTC staining method was used to detect the cerebral infarction volume. Wet and dry weights were used to measure brain water content. RT-PCR analysis was used to determine the activity of NF-κBmRNA expression. Results: Compared with control group,ischemia-reperfusion group's dosing volume of infarction rats at each time point,the brain water content and the expression of NF-κBmRNA were significantly increased( P〈0. 01). Compared with ischemia-reperfusion group,cerebral infarction volume,brain water content and the expression of NF-κBmRNA in each drug group decreased significantly at each time point( P〈0. 01). Compared to ginaton group,cerebral infarction volume,brain water content and the expression of NF-κBmRNA in Shida Lazhi Pill groups reduced or increased at each time point,especially in high dose group. The difference was statistically significant( P〈0. 01). Conclusion: Shida Lazhi Pill not only can reduce infarct volume and brain water content but also can obviously decrease the expression of NF-κB in brain tissue,inhibiting inflammatory reaction. The role of neural protection mechanism may be related to inhibiting inflammatory reaction.
