摘要
目的探讨羟基酪醇和橄榄苦苷在氧化应激致猪LLC-PK_1(p LLC-PK1)细胞损伤中的拮抗作用。方法用10~200μmol/L过氧化氢处理pLLC-PK_1细胞,筛选最佳的处理时间与浓度,建立LLC-PK_1细胞过氧化氢氧化应激体外模型。LLC-PK1细胞分别与2、5和10μmol的羟基酪醇和橄榄苦苷孵育12 h,用100μmol/L的过氧化氢作用4 h,裂解LLC-PK_1细胞后,检测丙二醛(MDA)含量、超氧化物歧化酶(SOD)活力和谷胱甘肽过氧化物酶(GSH-Px)活力和脂肪酸组成变化。结果与过氧化氢模型组(100μmol/L)相比,在橄榄苦苷和羟基酪醇处理的LLC-PK_1细胞中,MDA表达量降低,SOD和GSH-Px活力显著升高,脂肪酸转化降低,差异均有统计学意义(P〈0.05,P〈0.01)。结论羟基酪醇和橄榄苦苷对过氧化氢诱导的LLC-PK_1细胞氧化应激具有拮抗作用。
Objective To explore the antagonism of hydroxytyrosol and oleuropein on porcine LLC-PK_1 cells caused by hydrogen peroxide(H_2O_2). Methods The in vitro oxidative stress model of the LLC-PK_1 cells was established based on the opitimized time and concentration of the H_2O_2 treatment, cell viability was tested by MTT assay. After the LLC-PK_1 cells were incubated with 2 μmol/L, 5 μmol/L and 10 μmol/L of hydroxytyrosol and oleuropein for 12 h,respectively,100 μmol/L of H2O2 was added into the treated LLC-PK_1 cells,incubating for 4 h. After the LLC-PK_1 cells were lysed,the MDA content,SOD activity,GSH-Px activity,and fatty acid transformation were detected,respectively. Results Compared with H_2O_2 model group,in the LLC-PK_1 cell groups treated by hydroxytyrosol and oleuropein,the MDA content decreased significantly,the activities of both SOD and GSH-Px exhibited a significant increase,and the fatty acid transformation showed a significant decrease.Conclusion Hydroxytyrosol and oleuropein have a potential preventive effect on LLC-PK_1 cells suffering from the damage by H_2O_2 oxidative stress.
