摘要 目的 :探讨microRNA-148b-3p(miR-148b-3p)对施万(Schwann)细胞增殖的影响及作用的靶基因。方法 :采用Edu染色法检测miR-148b-3p对原代培养的施万细胞增殖能力的影响;基因芯片及生物信息学分析筛选miR-148b-3p可能作用的靶基因;实时定量PCR检测施万细胞中过表达miR-148b-3p模拟物后靶基因的表达变化。结果 :miR-148b-3p能明显促进原代培养的施万细胞的增殖,miR-148b-3p过表达可抑制其靶基因Alcam、Cpd、Dedd、Nptx2、Phf20、Ptpn14和Stard 13的表达。结论 :miR-148b-3p有可能通过对增殖相关靶基因的负调控来影响施万细胞的增殖能力,从而促进周围神经损伤修复。 Objective: To investigate the target genes effect of microRNA-148b-3p(miR-148b-3p) in Schwann cell proliferation.Methods: Edu staining analyses were performed to examine the proliferation of primary cultured Schwann cells after transfecting miR-148b-3p mimics. Microarray analysis and bioinformatic prediction were used to choose potential target genes of miR-148b-3p. And the expressions of target genes after miR-148b-3p mimics transfection on Schwann cells were detected by quantitative real-time PCR. Results: miR-148b-3p could significantly promote the proliferation of primary cultured Schwann cells by suppressing the expression of its target genes(Alcam,Cpd,Dedd, Nptx2, Phf20, Ptpn14 and Stard13).Conclusion: miR-148b-3p could affect Schwann cell proliferation by negatively regulating its target genes involving in proliferation, and facilitate peripheral nerve regeneration.
