黄芩素对人结肠癌细胞增殖、凋亡、侵袭的影响及机制

Effects of baicalein on proliferation,apoptosis and invasion of human colorectal cancer cells

目的探讨黄芩素对人结肠癌细胞增殖、凋亡、侵袭的影响及其作用机制。方法取处于对数生长期的人结肠癌SW480细胞,随机分为对照组,25、50、100μmol/L黄芩素处理组,分别加入生理盐水,25、50、100μmol/L黄芩素处理,采用Ed U、MTT、流式细胞术及Transwell小室实验观察黄芩素对结肠癌细胞增殖、凋亡及侵袭的影响,并通过Western blotting法检测黄芩素干预后结肠癌细胞Bcl-2、Bax及VEGF蛋白表达。结果 25、50、100μmol/L黄芩素处理组SW480细胞增殖受到抑制,呈剂量、时间依赖性;25、50、100μmol/L黄芩素处理组SW480细胞凋亡率依次升高;25、50、100μmol/L黄芩素处理组SW480细胞的侵袭迁移能力降低;黄芩素干预后SW480细胞中Bcl-2、VEGF蛋白表达下调,Bax蛋白表达上调(P<0.05)。结论黄芩素抑制人结肠癌细胞的增殖及侵袭,并促进其凋亡,机制可能与下调Bcl-2、VEGF蛋白表达并上调Bax蛋白表达有关。

Objective To investigate the effects of baicalein on proliferation,apoptosis and invasion of human colorectal cancer cells and to explore the mechanism. Methods Human colorectal cancer cells SW480 in logarithmic phase were randomly divided into the control group,25,50 and 100 μmol / L baicalein treatment groups which were respectively treated with normal saline,25,50 and 100 μmol / L baicalein. The effects of baicalein on proliferation,apoptosis and invasion of SW480 cells were detected by Ed U,MTT,flow cytometry and Transwell test. The protein expression of Bcl-2,Bax and vascular endothelial growth factor（ VEGF） in SW480 cells after the treatment of baicalein was detected by Western blotting. Results In the 25,50 and 100 μmol / L baicalein treatment groups,the proliferation of SW480 cells was inhibited with a dose and time-dependent manner,the apoptosis rates of SW480 cells were gradually increased,and the invasion abilities of SW480 cells were gradually decreased. After the treatment of baicalein,the expression of Bcl-2 and VEGF in SW480 cells was down-regulated,and the expression of Bax was up-regulated（ P〈0. 05）. Conclusion Baicalein inhibits the proliferation and invasion of SW480 cells and promots the apoptosis,which might be related with the down-regulation of Bcl-2 and VEGF expression and the up-regulation of Bax expression.
