外源性H2S改善老年大鼠血管功能

摘要 目的探讨H:S对老年血管功能的影响。方法雄性sD大鼠30只,随机分为年轻组,老年大鼠对照组,老年大鼠NariS(H2S的供体)低(1μmol·kg^-1·d^-1)、中(5μmol·kg^-1·d^-1)、高(10μmol·kg^-1·d^-1)剂量组,每组6只。颈总动脉插管观察NariS对老年大鼠心率及动脉血压的影响,离体血管环灌流方法评估大鼠血管收缩及舒张功能,Masson染色方法观察血管形态学改变。结果与年轻组相比,老年大鼠动脉血压降低(P〈0.05),心率明显减慢,血管收缩舒张功能受损(P〈0.05),给予低、中、高剂量的NariS后,动脉血压均没有明显改善,但老年大鼠的心率明显增加(P〈0.05);NariS中剂量组能明显改善老年大鼠血管收缩舒张功能(P〈0.05)。与年轻组相比,老年大鼠血管胶原纤维增多,给予低、中、高剂量的NaHS后,血管周围胶原沉积有不同程度减少,NaHS中、高剂量组更为明显。结论适量的外源性H:S能够改善老年大鼠血管的功能。。 Objective To discuss the effect of exogenous hydrogen sulfide (H2 S) on vascular function in aging rats. Methods A total of 30 adult male SD rats were randomly divided to fige groups (n = 6) : a young group, a control group, as well as a low - dose NariS group ( 1 μmol·kg^-1·d^-1 ), a medium - dose NariS group (5 μmol·kg^-1·d^-1 ) and a high - dose NariS group ( 10 μmol·kg^-1·d^-1). The vascular function was evaluated through common carotid artery intu- bation and vascular ring perfusion in vitro. Morphological changes of the aorta were observed by Masson staining. Results Compared with the young group, aging rats presented decreased heart rates and arterial pressure, as well as impaired con- striction and dilation function (P 〈 0.05). NariS treatment at different doses produced no effect on arterial pressure, but improved heart rates (P 〈 0.05). The constriction and dilation functions were improved after administration with 5 μmol·kg^-1·d^-1 NariS (P 〈 0.05 ). Compared with the young group, fibrosis was observed in aging rats. After treatment with different doses of NariS, collagen deposition was reduced, especially in the medium - and high - dose Naris Groups. Conclusion A balanced amount of exogenous hydrogen sulfide can improve vascular function in aging rats.
