丹参酚酸B对糖皮质激素诱导骨丢失大鼠腰椎骨密度和骨微结构的影响

摘要 目的探讨丹参酚酸B对糖皮质激素诱导骨丢失大鼠腰椎骨密度和骨微结构的影响。方法 40只7月龄雌性SD大鼠随机等分为空白对照组(CON组),糖皮质激素模型组(GC组)、糖皮质激素+丹参酚酸B组(GC+SAB组)、糖皮质激素+骨化三醇组(GC+CALM),分别以生理盐水(5 mL·kg^(-1)·d^(-1)),泼尼松(6 mg·kg^(-1)·d^(-1))、泼尼松+丹参酚酸B(25 mg·kg^(-1)·d^(-1)、泼尼松+骨化三醇(0.045μg·kg^(-1)·d^(-1))灌胃14周。取第五腰椎进行Micro-CT扫描并对松质骨进行三维重建,观察骨微结构并检测骨密度等参数。结果与CON组比较,GC组大鼠腰椎骨密度、骨体积分数、骨小梁数目及厚度分别下降13%、21%、7%、13%,而结构模型指数上升174%。与GC组比较,GC+SAB组大鼠腰椎骨密度、骨体积分数、骨小梁数目及厚度分别上升18%,30%,16%,12%,而结构模型指数、骨小梁分离度分别下降227%,15%。结论丹参酚酸B(25 mg·kg^(-1)·d^(-1))可预防糖皮质激素诱导骨丢失大鼠腰椎骨密度下降和骨微结构异常。 Objective To investigate the effect of salvianolic acid B(SAB) on bone mineral density(BMD) and microstructure of lumbar vertebras in rats with bone loss induced by glucocorticoid.Methods Forty 7-month-old female SD rats were evenly randomized to control group,glucocorticoid(GC) group,GC + SAB group,GC + calcitriol(CAL) group,each receiving intragastric infusion of normal saline(5 mL·kg^-1·d^-1,prednisone(6 mg·kg^-1·d^-1,prednisone + SAB(25 mg·kg^-1·d^-1and prednisone +calcitriol(0.045μg·kg^-1·d^-1 for 14 days,respectively.The fifth lumbar vertebra was excised for Micro-CT scanning,three-dimensional reconstruction of trabecular bone,and observation of bone microstructure and BMD.Results Compared with CON group,BMD,bone volume fraction,and trabecular number and thickness were respectively decreased by13%,21%,7%,and 13%,while structure model index was increased by 174%in GC group.Compared with GC group,BMD,bone volume fraction,and trabecular number and thickness were increased by 18%,30%,16%,and 12%,whereas structure model index and trabecular separation were decreased by 227% and 15%in GC+SAB group,respectively.Conclusion SAB(25mg·kg^-1·d^-1 can prevent the decreased BMD and abnormal bone microstructure of lumbar vertebras in rats with bone loss induced by glucocorticoid.
