丹参酮与DNA相互作用的电化学研究

摘要 目的研究丹参酮Ⅰ、二氢丹参酮Ⅰ、丹参酮ⅡA和隐丹参酮与DNA的相互作用。方法采用电化学方法计算结合数,结合常数,并推测结合方式,用紫外光谱进一步验证结合方式。结果每4摩尔丹参酮Ⅰ、二氢丹参酮Ⅰ或丹参酮ⅡA可与1摩尔DNA碱基对形成复合物,而每1摩尔隐丹参酮可与1摩尔DNA碱基对结合形成复合物。它们与DNA的结合能力由强到弱分别为:丹参酮Ⅰ、二氢丹参酮Ⅰ、丹参酮ⅡA和隐丹参酮。结合方式均为静电作用。结论丹参酮的共轭体系越大与DNA结合越牢固,并且不共面的甲基还具有空间位阻效应,降低丹参酮与DNA的结合能力。 Objective To study the interaction between tanshinones including tanshinone I, dihydrotanshinone I, tanshinone IIA and cryptotanshinone and DNA. Methods The binding number and constant and bonding form were calculated by electrochemistry and validated by UV spectrophotometry. Results Four moles of tanshinone I, dihydrotanshinone I or tanshinone IIA, and 1 mole of cryptotanshinone were combined with 1 mole of DNA base pair via electrostatic interaction. The binding capacity was tanshinone I, dihydrotanshinone I, tanshinone IIA and cryptotanshinone in descending order. ConclusionThe conjugated system of tanshinones is positively correlated with DNA-binding capacity. The steric-hinerance effect of the noncoplanar methyls can also reduce the DNA-binding capacity.
