摘要
为研究开辟香蕉资源综合利用新途径,以香蕉皮及麸皮为主要原料,黑曲霉CICC40493为发酵菌种,采用单因素和正交试验,研究确定固态发酵果胶酶的基础培养基配比。结果显示,果胶酶最适基础培养基为:香蕉皮渣3g、麸皮7g、硫酸铵0.45g、水15m L;影响酶活力的主次顺序为:麸皮和香蕉皮渣配比>硫酸铵添加量>去离子水添加量,最佳配比的培养基在自然p H值,装瓶量10g干料/250m L三角瓶,接种量1.0m L的条件下,28℃恒温培养72h,获得的果胶酶总活力为10853.54U。试验结果表明利用香蕉皮接种黑曲霉CICC40493固态发酵生产果胶酶是可行的,为开辟香蕉资源综合利用新途径的研究提供数据支持。
To prepare banana peel and wheat bran as main raw materials, Aspergillus Niger CICC40493 as fermentation, and then to adopt single factor and orthogonal experiments determine mixture ratio of solid-state fermentation pectinase basic culture medium. The result showed that the optimum culture medium for pectinase was as follow: banana peel 3g, bran 7g, ammonium sulfate 0.45 g, deionized water 15 mL;The primary and secondary sequence of the enzyme activity was the ratio of banana peel and wheat branaddition of ammonium sulfateaddition of deionized water, the optimal proportion of medium was fermented in the following conditions: natural p H, bottling capacity 10 g drier/250 mL triangle bottle, inoculation 1.0mL, 28℃ as fermentation temperature,and 72 hours as fermentation time, then, experimenter would get the total activity pectinase was 10853.54 U. The result showed that producing pectinase by the banana peel inoculated with solid state fermentation of Aspergillus niger CICC40493 was feasible.
