蓝莓对非酒精性脂肪性肝炎大鼠肝组织γ-谷氨酰半胱氨酸合成酶催化亚基蛋白表达的影响 被引量:3

摘要 目的:研究非酒精性脂肪性肝炎(NASH)大鼠γ-谷氨酰半胱氨酸合成酶催化亚基(GCLC)表达的变化及蓝莓对其表达的影响。方法:40只健康SD大鼠随机分为4组:正常组、蓝莓组、异甘草酸镁组、模型组,每组10只。正常组大鼠饲喂正常饲料,其余各组大鼠饲喂高脂饲料。蓝莓组大鼠给予蓝莓原浆(15ml·kg^(-1)·d^(-1))灌胃;异甘草酸镁组大鼠给予异甘草酸镁溶液(15ml·kg^(-1)·d^(-1))灌胃;其余组大鼠给予等量生理盐水灌胃。12周后处死大鼠,HE染色观察各组大鼠肝脏病理变化;全自动生化分析仪检测大鼠血清中丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)、总胆固醇(TC)、甘油三酯(TG)、含量;酶联免疫法检测肝组织中还原性谷胱甘肽(GSH)、中丙二醛(MDA)、超氧化物歧化酶(SOD)的含量;免疫印迹法检测大鼠肝组织中γ-谷氨酰半胱氨酸合成酶(γ-GCS)催化亚基(《GCLC)的蛋白表达。结果:异甘草酸镁组、蓝莓组大鼠肝损害较模型组明显减轻,血清ALT、AST、TC、TG含量明显降低(P<0.05),肝组织中MDA含量明显降低(P<0.05),GSH含量、SOD活性明显增加(P<0.05);模型组大鼠GCLC蛋白表达较正常组增加,但无明显差异(P>0.05),蓝莓组和异甘草酸镁组则增加(P<0.05)。结论:蓝莓对NASH大鼠肝组织具有保护作用,机制可能与上调GCLC蛋白表达有关。 Objective:To investigate the effects of blueberry on GCLC protein expression in liver tissue ofrats with nonalcoholic fatty liver disease.Methods;40 healthy SD rats were randomly divided into normalcontrol group,blueberry group,magnesium isoglycyrrhizinate group and model group,10 rats in each group.The rats in normal control group were given normal diet,those in other groups were all given high fat diet.At the same time,blueberry group were intragastrically given blueberry juice(15ml ·kg^-1 · d^-1),magnesium isoglycyrrhizinate group were gavaged magnesium isoglycyrrhizinate solution(15ml · kg^-1 · d^-1),the remaining rats were given saline gavage.The rats were killed after 12 weeks,the pathological changes of the liver were observed by HE staining;the level of alanine aminotransferase(ALT).aspartate aminotransferase(AST),triglyceride(TG) and total cholesterol(TC) in serum were detected by automatic biochemical analyze;The hepatic MDA contents,GSH levels and SOD activity were quantified by ELISA.The expression of GCLC in liver tissue was detected by Western blot.Results;Compared to model group,blueberry group and magnesium isoglycyrrhizinate group showed decreased in liver damage,ALT,AST,TG,MDA and TClevels(P〈0.05).while GSH contents and SOD activity increased(P〈0.05).Compared to normal group,the expression of GCLC levels in model group had no significant increased(P〈0.05),while blueberry group and magnesium isoglycyrrhizinate group increased(P〈0.05).Conclusion:Blueberry has protective effect on liver tissue of NASH rats.which may be related to up regulate the GCLC protein expression.
