高效液相色谱法测定常见食物中β-胡萝卜素含量 被引量:11

Determination of β-Carotene in Common Foods by High Performance Liquid Chromatography

摘要 采用高效液相色谱法测定300多种市售主食食材中β-胡萝卜素含量,具体条件:色谱柱为Agilent Zobrax SB-C18(4.6 mm×250 mm,5μm),流动相条件为:含0.4 g/L抗坏血酸的甲醇/乙腈/三氯甲烷(84/12/4)溶液(避光储存);流速1.5 m L/min;柱温:30℃;检测波长:450 nm,进样体积10μL,外标法定量。结果表明该方法在0.50020.0μg/mL质量浓度范围内线性良好,相关性系数R=0.999 9,RSD=2.4%,方法检出限为0.2 mg/kg,回收率为78.7%96.4%。该方法整体操作简单,线性范围好,结果可靠。该方法通过高效液相色谱法完成了300多种深圳市售主食材中β-胡萝卜素含量测定,首次建立β-胡萝卜素含量数据库,并与我国现有的胡萝卜素含量数据库和美国β-胡萝卜素含量数据库进行了比较。 Zobrax SB-C18(4.6 mm×150 mm, 5 μm), mobile phase: 0.4 g/L ascorbic acid was dissolved in methanol/ acetonitrile/chloroform solution with volume ratio of 84/12/4(keep in darken place and avoid light); Mobile phase rate: 1.5 mL /min; oven temperature: 30 ℃; Measuring wavelength: 450 nm; Injection volume: 10 μL, external standard method was used for qualification. The result showed that this method had good linearity in the concentration range of 0.50020.0 μg /mL, the correlation coefficient(R) was 0.999 9, RSD was 2.4%, the detection limit was 0.2 mg/kg and recovery was in the range of 78.7%96.4%. This method had the charecters of easy operation, good linearity and reliable results. By employng this method, 300 kinds of foodstuffs that consumed in Shenzhen were sampled to test β-carotene concentration, based on the result obtained in our research, β-carotene content database was built and comparation with China existing β-carotene database and America β-carotene database were also made.
