摘要
目的研究茯苓、桂枝单味药和茯苓桂枝复方对慢性心肌缺血大鼠的治疗作用。方法 60只SPF级大鼠分为正常组、模型组、茯苓组、桂枝组、茯苓桂枝组和心得安组,共6组,慢性心肌缺血造模成功后,正常组和模型组灌服生理盐水,其余各组分别灌服茯苓煎液、桂枝煎液、茯苓桂枝煎液,以及心得安灌胃30d。记录造模前后和给药后心电图ST段变化,测血清中乳酸脱氢酶(LDH),肌酸激酶(CK)和肌酸激酶同工酶(CK-MB)酶水平;取各组动物的心脏组织,观察组织病理变化。结果灌胃后,与模型组比较,茯苓组、桂枝组、茯苓桂枝组、心得安组ST段电压降低,有显著性差异(P<0.01)。与模型组比较,茯苓桂枝组能有效降低血清中CK、CK-MB、LDH含量,有显著性差异(P<0.01)。病理组织观察结果表明茯苓桂枝组组织切片较模型组有较大改善,纹理清晰,肌纤维排列整齐等,心得安组与正常组相比无显著差异。结论茯苓-桂枝药对对大鼠慢性心肌缺血有保护心肌作用,并且茯苓桂枝复方药较单味药保护作用更好。
Objective To investigate the effects of the chronic myocardial ischemia of rats by gavage of Fuing,Guizhi respectively and Fuling- Guizhi hebal compatibility. Methods 60 SPF rats were divided into 6 groups randomly: normal group,Propranolol group,model group,Fuling group,Guizhi group,Fuling- Guizhi group. After successfully buliding the models of Chronic myocardial ischemia,rats of normal group and model group were given saline solution once a day for 2 weeks simultaneously. Rats of Propranolol group were given Propranolol,the rest groups were given drugs for 30 days. After 30 days of drug intervention,changes of electrocardiogram ST segment,levels of lactate dehydrogenase( LDH),creatine kinase( CK),creatine kinase- MB( CK- MB) and HE staining of the heart tissue were observed. Results Compared with model group,ST segment voltage of Fuling group,Guizhi group,Fuling- Guizhi group,Propranolol group reduced,had significant difference( P〈0. 01). Compared with model group,Fuling- Guizhi group effectively reduced the content of CK,CK-MB,LDH in serum( P〈0. 01). Histopathological results showed Fuling- Guizhi group,compared with the model group,was more effectively in improving: degrees of texture clearity of tissue sections and reducing cell focal necrosis. Compared with the normal group,Propranolol group showed no significant difference. Conclusion Fuling- Guizhi herbal compatibility has protective effects on chronic myocardial ischemia,and is more effective than giving Fuling or Guizhi respectively.
