三七总皂苷、红景天苷、黄芪有效组分对心肌梗死大鼠模型骨髓间充质干细胞动员作用研究 被引量:14

Effect of Notoginseng Radix et Rhizoma Saponins,Salidroside and Effective Components of Astragali Radix in Mobilizing Bone Marrow Mesenchymal Stem Cells in Rat Myocardial Infarction Model

摘要 目的:探讨三七总皂苷、红景天苷、黄芪有效组分对心肌梗死大鼠模型骨髓间充质干细胞(bone mesenchymal stem cells,BMSCs)动员作用。方法:选取SPF级SD大鼠,参考Olivette方法建立急性心肌梗死模型,造模成功大鼠随机分为活血组、益气组、益气活血组、模型组和正常组,各组再随机按1,3,7,14,28 d时间点再进行分组。活血组8 mg·kg-1血塞通软胶囊、益气组5 mg·kg-1黄芪颗粒、益气活血组7 mg·kg-1诺迪康胶囊,模型组予生理盐水ig,正常组不给药,正常组和模型组以等量生理盐水ig,1次/d,共28 d。流式细胞仪检测不同时间点血液和骨髓中CD54,CD106,CD105,CD117阳性细胞百分数,酶联免疫吸附测定(ELISA)法检测不同时间点血液和骨髓中干细胞因子(SCF)变化。结果:模型组外周血和骨髓中CD105,CD117,SCF从1 d开始升高,14 d到高峰,其后外周血和骨髓中CD105,CD117,SCF含量降低;活血组、益气活血组、益气组外周血和骨髓中CD105,CD117,SCF含量上升优于模型组(P<0.05)。活血组CD105,CD117,SCF上升优于益气活血组、益气组。模型组外周血和骨髓中CD54,CD106在1 d到高峰,其后逐渐降低,活血组外周血中的CD54,CD106降低明显较模型组相比(P<0.05)。益气组与模型组相比变化不明显,但活血化瘀方药降低CD54,CD106最明显。结论:三七总皂苷、红景天疳、黄芪有效组分均有动员骨髓间充质干细胞作用,但活血化瘀方药动员效果明显。 Objective: To investigate the effect of Notoginseng Radix et Rhizoma saponins,salidroside and effective components of Astragali Radix in mobilizing bone marrow mesenchymal stem cells in the rat model of myocardial infarction. Method: The SD rat model of myocardial infarction was established by Olivette method.The successfully modeled rats were randomly divided into: Huoxue group,Yiqi group,Yiqi and Huoxue group,model group and normal group,and then subdivided according to time points at 1,3,7,14,28 d. Huoxue group was given 8 mg·kg- 1of Xuesaitong soft capsule. Yiqi group were treated with 5 mg·kg- 1of Huangqi granule. Yiqi and Huoxue group was given 7 mg·kg- 1of Nuodikang capsule. Model group was given normal saline. Normal group received no drug. Normal group and model group were administered with equivalent normal saline by gavage once a day for 28 days. The positive cells percentage of CD54,CD106,CD105,CD117 in peripheral blood and bone marrow were detected by flow cytometry at different time points,and ELISA method was used to detect the expression of stem cell factor( SCF) in peripheral blood and bone marrow at different time points. Result: SCF level and positive cells percentage of CD105,CD117 content increased in model group since the first day,reached the peak on the 14 th day,and then decreased gradually. But the increasing trend was superior in Huoxue group,Yiqi group,Yiqi and Huoxue group( P 〈 0. 05),particularly Huoxue group was the best. The prescription of promoting blood circulation to remove blood stasis showed an obvious mobilizing effect. Positive cells percentage of CD54,CD106 in peripheral blood and bone marrow in model group reached the peak on the first day,and then decreased gradually,but Huoxue group showed a superior decreasing trend in CD54,CD106 than model group in peripheral blood( P 〈 0. 05). Compared with model group,CD54 and CD106 in bone marrow had no significant change. Conclusion: Notoginseng Radix et Rhizoma saponins,salidroside and effective components of Astragali Radix can mobilize bone marrow mesenchymal stem cells,and the mobilizing effect of the prescription of promoting blood circulation to remove blood stasis is the most obvious.
