摘要
为开发具有改善养殖水体富营养化作用的有益菌,从江苏连云港滩涂海水池塘分离到1个菌株,命名为HYHB-4,对其进行形态学特征及生理生化特性鉴定,并分析其16S rRNA序列,结果表明,分离菌株为蜡状芽孢杆菌(Bacillus cereus)。研究了分离菌株对虾蟹混养围塘水环境中氮磷的纠正效应,结果显示,HYHB-4菌株中度嗜盐、嗜冷,最佳生长温度为25℃,超过35℃停止生长;在3%-5%Nacl中生长得率最高;菌株HYHB-4对虾蟹混养围塘水环境中的氮磷具有显著的利用转化和降解作用,其中亚硝态氮(NO2-N)下降幅度最大,施用HYHB-4制剂5 d后,氨态氮(NH4-N)、硝态氮(NO3-N)、NO2-N质量浓度比起始水平分别下降77.19%、52.95%、85.27%;总磷(TP)和磷酸盐(PO4-P)比起始水平分别下降84.40%、84.67%。综上,分离的蜡状芽孢杆菌HYHB菌株-4对虾蟹混养围塘水环境中的氮磷具有纠正效应。
In order to isolate and identificate probiotic which can inprove the eutrophication effects of aquaculture water,one strain was isolated from the seawater pond of Lianyungang in Jiangsu province,the strain was named by HYHB-4. From morphological characteristics and physiological and biochemical characteristics,combined with the 16 S rRNA sequence analysis,the strain preliminarily identified as Bacillus cereus. The corrective effect of nitrogen and phosphorus of the strain to the water environment in shrimp and crab polyculture pond were investigated. The results showed that the strain HYHB-4 moderate halophilic and psychrophilic,the optimum growth temperature was 25 ℃,and it would stop growing exceeding 35 ℃. The growth was highest in 3% —5% salinity. The strain HYHB-4 had significant transformation and degradation function to the nitrogen and phosphoru of water environment in shrimp and crab polyculture pond. The NO2-N had the largest decline. After using five days,the concentrate of NH4-N,NO3-N,NO2-N respectively decreased by 77. 19%,52. 95%,85. 27% than the starting level. The TP and PO4-P respectively decreased by 84. 40%,84. 67% than the starting level. The experiment confirmed that the strain HYHB-4 had a significant corrective effect on the nitrogen and phosphoru of water environment in shrimp and crab polyculture pond.
