Expression of Nicotinic Acetylcholine Receptors on Primary Culture of Rat Astrocytes

摘要 该文研究了尼古丁型乙酰胆碱受体(nicotinic acetylcholine receptors,n ACh Rs)不同亚单位在大鼠大脑星形胶质细胞的表达情况。分离新生大鼠大脑皮质培养原代星形胶质细胞并纯化、传代鉴定后,运用RT-PCR方法、蛋白质印迹法检测正常大鼠大脑皮质星形胶质细胞n ACh Rs不同亚单位的表达情况。结果显示,α2、α3、α4、α7、β2等n ACh Rs亚单位在星形胶质细胞均有表达,而β3 n ACh Rs亚单位在星形胶质细胞没有表达,为深入研究星形胶质细胞的功能提供了可能。 This work was aimed to investigate the expression of different subunits of the nicotinic acetylcholine receptors(nAChRs) on primary culture of rat astrocytes. Primary culture of rat astrocytes were prepared from the cortex and hippocampus of the brains of newborn Sprague-Dawley(SD) rats. The m RNA and protein levels of individual n ACh R subunits in primary cultured astrocytes of rat were examined by RT-PCR and Western blot. The α2, α3, α4, α7 and β2 n ACh R subunits were found in the primary cultured astrocytes while the β3 n ACh R subunit was not presented, which provide some evidence for further function study of astrocytic n ACh Rs.
