摘要
目的通过检测中药漏芦乙酸乙酯提取物(ethyl acetate extract,EA)对口腔鳞癌细胞株SCC15中转录因子Ets-1及过氧化物还原酶家族成员Prx1的表达水平的影响,初步探讨漏芦对口腔鳞癌细胞氧化应激作用的分子机制,为指导口腔癌临床用药提供新的思路。方法 50μg/ml漏芦EA处理口腔鳞癌细胞株SCC15 48h后,采用荧光定量PCR和Western Blot,检测药物处理组和对照组中Ets-1和Prx1的mRNA与蛋白表达情况。结果倒置显微镜下观察,50μg/ml漏芦EA处理SCC15细胞48h后,细胞生长速度较对照组减慢,细胞形态变圆,部分出现皱缩变圆甚至脱落。与对照组相比,药物处理组中Ets-1和Prx1的mRNA相对表达量显著降低(P=0.002,P=0.025),蛋白表达水平显著降低(P=0.030,P=0.042),差异有统计学意义。结论漏芦可能通过抑制氧化应激相关蛋白Ets-1和Prx1表达来抑制口腔癌细胞的生长。
Objective To investigate the effect of Radix rhapontici on the expression of transcription factor Ets-1and Prx1 in oral cancer. Methods SCC 15 cells were treated with 50μg / ml EA extract of Radix rhapontic for 48 h. The cell morphology was observed under inverted microscope,mRNA and protein expression of Ets-1 and Prx1 were detected by quantitative real-time PCR and western blot,respectively. Results SCC15 cells grew slowly and cell morphology changed from polygonal to round shape under inverted microscope after the cells were treated with EA extract of Radix rhapontic for48 h. Compared with control group,the EA extract of Radix rhapontic inhibited mRNA and protein expression of Ets-1 and Prx1 in SCC15 cells( P〈 0. 05). Conclusion EA extract of Radix rhapontic may inhibit the cell proliferation via regulating Ets1 and Prx1 expression in oral cancer.
