Blyscan法测定黏多糖贮积症患者尿液中黏多糖含量的临床意义

The clinical significance of quantitative measuring urinary glycosaminoglycans by Blyscan method in mucopolysaccharidosis patients

目的研究Blyscan检测法对黏多糖贮积症(MPS)患者尿液中黏多糖(GAG)含量测定的效能及临床意义。方法对Blyscan检测法进行日内精密度、日间精密度、时间稳定性、加样回收率的检测,并对2013年10月—2015年6月解放军总医院儿童医学中心已确诊的25例MPS患者(MPS组)尿液和20例正常健康人(健康对照组)的尿液中的GAGs进行定量检测。结果 Blyscan检测法测量低值和高值标准溶液的日内精密度CV值分别为0.96%和1.32%,日间精密CV值分别为2.33%和1.85%。Blyscan法的10、20 mg/L的回收率为(97.04±3.61)%、(100.32±5.62)%,DMB-Tris回收率为(95.01±1.62)%、(103.9±2.81)%,两者差异无统计学意义(P>0.05)。但与DMBTris比色法相比,Blyscan检测法结果稳定性较好(P<0.05)。用Blyscan法检测MPS组尿GAG/Cr比值为(65.36±21.96)mg/mmol,健康对照组为(12.32±1.75)mg/mmol,前者较后者有显著升高(P<0.05)。结论 Blyscan检测法是一种准确度高、稳定性好、操作简便的GAGs检测方法,适用于临床对GAGs含量的定量检测。

Objective To study the efficiency of the Blyscan method for measuring urinary glycosaminoglyc.ans（GAGs） in mucopolysaccharidosis（MPS） patients and its clinical significance.Methods The intra-day and inter-day precisions,temporal stability,average recovery were detected by Blyscan method in the study.The urinary GAGs of 25 cases of MPS patients（MPS group） diagnosed by pediatric department and 20 health cases（normal control group） from October 2013 to June 2015 were detected by Blyscan method.Results The CV values of the intra-day precisions for detecting low-value and high-value standard solution by Blyscan method were 0.96%and 1.32%,and the CV values of inter-day precisions of them were 2.33%and 1.85%,respectively.And what＇s more,the recoveries of 10 and 20 mg/L detected by Blyscan method and DMB-Tris method were（97.04 ±3.61）%,（100.32 ±5.62）%and（95.01 ± 1.62）%,（103.9 ±2.81）%,respectively,without significantly difference（P〉0.05）.However,the stability of Blyscan method was much more stable than DMB-Tris method（P〈0.05）.Compared the value of urinary GAG/Cr between the MPS patients and control,the former was significant higher than the latter[（65.36 ±21.96）mg/mmol vs.（12.32 ± 1.75）mg/mmol,（P〈0.05）].Conclusion It proved that the Blyscan method,which is suitable for the clinical quantitative detection of GAGs content,was a reliable,stable and simple GAGs quantitative assay method.
