摘要
目的:观察柴芩通淋片对慢性非细菌性前列腺炎大鼠模型的作用,探讨其发挥作用的可能机制。方法:制作慢性非细菌性前列腺炎大鼠模型,分组造模14 d后给予柴芩通淋片混悬液灌胃3周,空白对照组及模型组给予相应剂量的生理盐水灌胃,灌胃结束后取前列腺组织及前列腺液。前列腺组织石蜡包埋切片行HE染色,观察前列腺组织病变程度,免疫组化法观察TNF-α、IL-10阳性表达细胞数;前列腺液白细胞计数评级。结果:大体观察显示模型组大鼠前列腺显著充血和水肿,镜下HE染色显示实质内腺泡萎缩、腺腔变小,间质及腺腔内大量单个核细胞浸润,结缔组织增生,呈现明显的慢性炎症表现;前列腺液白细胞数量增多。用药后柴芩通淋片高、中、低剂量组大鼠前列腺炎性病变及纤维化程度得到较好的改善,前列腺液白细胞数量显著减少。模型组大鼠前列腺组织中TNF-α阳性细胞数显著增多,IL-10阳性细胞数显著下降。柴芩通淋片高、中、低剂量组大鼠TNF-α阳性表达细胞数低于模型组,柴芩通淋片高剂量组IL-10阳性细胞数高于模型组。结论:柴芩通淋片能够显著改善慢性非细菌性前列腺炎模型大鼠的炎症病理改变,其作用机制可能通过降低促炎性细胞因子TNF-α的表达及促进抗炎性细胞因子IL-10表达,与调节前列腺组织炎症反应有关。
Objectives: To investigate the effects of Cha!qinTonglin Plan (柴芩通淋片, CQTLP) on chronic non-bacterial prostatitis (CNP) rats model, and to explore the underlying mechanisms. Methods: The CNP rats model were made by injecting complete Freund's adjuvant. Fourteen days after the injection, suspension of CQTLP was intragastrie administrated to rats for three weeks respectively, and the same volume of saline was intragastrie administrated to rats in the control group and model group. Leukocytes in the expressed prostatic secretion were counted to evaluate the differences among groups. Prostate was derived and paraffin-embedded to detect pathological changes with HE staining. S-P immunohistochemieal staining was used to investigate the expression of cytokine TNF-α and IL-10. Results: Significant hyperemia and edema were found in prastates from the model rats, microscopic observation of paraffin sections with HE stainning showed atrophic glands with prostate glandular cavity shranking. Numerous monocytes were found in both connective tissue and gland lumina. CQTLP oral treatment significantly alleviated the lesions caused by injection in the prostate and significantly reduced the count of leukocyte in the expressed prostatic secretion. The expression of TNF-α-positive cells of model group rat prostate tissue sections was significantly increased compared with the control group, while IL-10-positive cells was significantly decreased. Conclusion: CQTLP oral treatment significantly alleviated the lesions caused by injection in the prostate. The underlying mechanisms could probably relate with the reduction of expression of proinflammatory cytokine such as TNF-α and the increased expression of anti-inflammatory factor of IL-10.
