摘要
杜仲是世界上唯一的硬橡胶树种,是能够替代三叶橡胶的最具开发潜力的胶源植物。本研究以杜仲胶合成下游途径Ref基因为研究对象,运用Codon W和CUSPS等研究Ref基因的密码子组成特点及使用偏好性。研究结果表明,杜仲Ref基因的GC含量为49%,密码子第三位碱基的GC含量为59%;杜仲Ref基因有28个偏好使用的密码子,其中约60%(17个)的密码子以G/C结尾;与大肠杆菌、酵母、拟南芥和烟草的基因组密码子使用偏好性比较,杜仲与拟南芥基因组密码子的使用偏性更为接近,拟南芥更适合作为Ref转基因的受体。本研究为预测杜仲Ref基因的异源表达,适宜表达系统的选择以及Ref基因密码子的改造以提高其在宿主中的表达,进行有效的基因功能验证提供理论参考。
Eucommia ulmoides is the only species producing hardy-rubber and the most potential rubber resource to replace Hevea brasiliensis. In this article, we studied the condon composition and usage bias of Ref gene using Codon W and CUSPS softwares. Refis one of the genes participating in the rubber biosynthase of Eucommia ulmoides. Our study showed that the GC content of Ref gene is 49%. The GC content in the third position is 59%. There are 28 codons used more often in the Ref gene, 17 of which ending with G or C. Compared with the codon usage bias of the E.coli, A.thaliana, N.tabacum and S.cerevisiae genome, we found that Eucommia is closer to N.tabacum. So N.tabacum is a better acceptor for Ref transgenosis. This research offers a theoretical reference for predicting heterogeneous expression, acceptor selection, codon transformation and so on.
