摘要
目的 建立一种与人类发病相似的胰岛素抵抗性2 型糖尿病大鼠模型.方法 选取SD 大鼠30 只,随机分为正常组和实验组,各15 只,正常组整个实验过程中灌胃生理盐水2ml/d.实验组以灌胃高脂高糖乳剂2ml/d 稳定饲养8 周后,每3 天腹腔注射小剂量STZ 15mg/kg4次诱发2 型糖尿病大鼠模型后,检测空腹血糖、糖耐量、胰岛素、胰岛素敏感指数等糖代谢指标.结果 与正常组相比,实验组空腹血糖,糖耐量实验(OGTT),胰岛素敏感指数(ISI)功等糖代谢指标都有显著差异(P〈0.05).结论 该模型具有糖代谢紊乱,其模型可视为研究人类胰岛素抵抗性2 型糖尿病理想的动物模型.
Objective To establishment of a similar disease in humans insulin resistancein type 2 diabetic rat model.Methods Select the SDrats 30 were randomly divided into normal and experimental groups, each 15, the normal group throughout the experiment intragastric saline 2ml / d. Theexperimental group received intragastric fat emulsion 2ml / d stable for 8 weeks, every 3 days by intraperitoneal injection of STZ 15mg/kg4 induced type 2diabetic rat model, respectively, using optical microscopy and transmission electron microscopy the morphology of the pancreatic tissue changes andultrastructural changes and fasting glucose, impaired glucose tolerance, insulin, insulin sensitivity index and glucose metabolism indicators.ResultsCompared with normal group, the experimental group, fasting blood glucose, the glucose metabolism of glucose tolerance test (OGTT), insulin sensitivityindex (ISI), power and other indicators have a significant difference (P 〈0.05), compared with the normal group, the damage to the pancreas of theexperimental grouppathological changes in a significant increase in ultrastructural changes of thedifferences are significant. Conclusion The model has adisorder of glucose metabolism, and pancreatic tissue morphology and ultrastructure of the variation of the model can be regarded as the study of humaninsulin resistance in type 2 diabetes ideal animal model.
