建立比格犬颈动脉粥样硬化狭窄模型 被引量:2

Establish Beagles'carotid atherosclerotic stenosis model.

摘要 目的 探索通过氮气干燥结合球囊损伤比格犬颈动脉内皮细胞,辅以高脂饮食,观察能否建立比格犬颈动脉粥样硬化狭窄模型.方法 19 只健康雄性比格犬随机分为2 组,A 组(假手术组 n=4)和B 组(动脉粥样硬化狭窄造模组 n=15).A 组仅分离右侧颈总动脉但不损伤;B 组用氮气干燥结合球囊损伤右侧颈总动脉内皮细胞,两组均高脂饮食饲养2 个月.行颈部血管彩超和数字减影血管造影(Digitalsubtraction angiography,DSA)检查,对比术前、术后变化,观察比格犬颈动脉狭窄和粥样硬化斑块形成情况.用SPSS17.0 软件包进行t 检验,以P〈0.05 视为差异有统计学意义.结果 造模后2 个月颈部血管彩超结果显示A 组颈动脉内膜光滑,无狭窄和斑块形成;B 组造模侧颈动脉可见不同程度的狭窄和斑块形成.DSA 检测结果显示:B 组狭窄率显著大于A 组(P=0.000).结论 通过氮气干燥结合球囊损伤比格犬颈动脉内皮细胞,辅以高脂饮食,能够建立比格犬颈动脉粥样硬化狭窄模型. Objective To explore whether the models of carotid atherosclerotic stenosis in Beagles can be established by the way of ballooninjury and nitrogen drying of endothelial cells in their carotid arteries with high-fat diet .Methods 19 healthy beagles in male were randomly divided into2 groups, group A (sham group, n = 4) and group B (atherosclerotic group n = 15). Carotid artery of right was only separated in group A ,not injuried ;group B was injured by the way of balloonjury and nitrogen drying of endothelial cells in their carotid arteries, both of them supported by high-fat diet.Vascular ultrasound and DSA was used to observed the carotid artery stenosis and atherosclerotic plaque formation two months after modeling. ResultsCarotid intima of group A is smooth, which show by results of the vascular ultrasound , without stenosis and plaque formation; Carotid arteries of group Bcan be seen varying of degrees of stenosis and plaque formation. DSA results show group B is much larger than group A in stenosis rate(P〈0.005).Conclusion The model of carotid atherosclerotic stenosis in Beagles can be established by the way of balloon injury and nitrogen drying ofendothelial cells in their carotid arteries with high-fat diet .
