摘要
目的观察芎芷地龙汤对偏头痛模型大鼠脑组织c-fos和降钙素基因相关肽(CGRP)蛋白表达的影响,探讨芎芷地龙汤对偏头痛的作用机制。方法将健康雄性SD大鼠48只,随机分为生理盐水组、模型组、舒马普坦组、中药治疗组,每组12只,给药干预后,皮下注射硝酸甘油注射剂10 mg/kg制作偏头痛动物模型,在皮下注射药物后4 h、6 h两个时间点取材,免疫组化法测定三叉神经脊束核和中脑导水管周围灰质c-fos和CGRP免疫阳性细胞表达情况。结果三叉神经脊束核阳性细胞表达:中药治疗组cfos、CGRP阳性细胞数在4 h和6 h两个时间点均较模型组显著减少(P<0.01);导水管周围灰质阳性细胞表达:中药组c-fos、CGRP阳性细胞数在4 h和6 h两个时间点与模型组比较均显著性减少(P<0.01)。结论芎芷地龙汤可减少NTG诱发的三叉神经脊束核c-fos免疫阳性细胞表达,进而减弱三叉神经血管系统的激活,也可减弱中脑导水管周围灰质c-fos免疫阳性细胞表达,阻止伤害性信息的进一步传递;可减少三叉神经脊束核和中脑导水管周围灰质CGRP阳性细胞的表达。
Objective To observe the infcuence of Xiongzhi Dilong decoction( XDD) on the expressions of c-fos and calcitonin gene-related peptide( CGRP) immunoreactive neurons in brain tissues of migraine model rats,and to explore its mechanism.Methods Forty-eight healthy male Sprague-Dawley( SD) rats were divided randomly and equally into 4 groups:saline group,migraine model group,sumatriptan group,XDD group.The rats were pretreated with sumatriptan,XDD or saline before received a subcutaneous injection of 10 mg / kg nitroglycerin or saline.After the rats received a subcutaneous injection 4 h and 6 h,the brain tissue samples were collected.Immunohistochemistry was applied to detect c-fos and CGRP immunoreactive neurons in trigeminal nucleus caudalis( TNC) and periaqueductal gray( PAG).Results Immunoreactive positive neurons in trigeminal nucleus caudalis:c-fos immunoreactive neurons and CGRP immunoreactive neurons in XDD group were decreased at both 4 h and 6 h,and there was statistical difference comparing with the migraine model group( P〈0.01).Immunoreactive positive neurons in periaqueductal gray( PAG) :c-fos immunoreactive neurons and CGRP immunoreactive neurons in XDD group were decreased at both 4 h and 6 h,and there was statistical difference comparing with the migraine model group( P〈0.01) as well.Conclusion XDD can reduce the NTG-induced c-fos immunoreactive neurons in trigeminal nucleus caudalis,and attenuate the activatin of trigeminovascular system,can reduce the CGRP immunoreactive neurons in periaqueductal gray as well,and inhibit nociceptive transmission.XDD can reduce the CGRP immunoreactive neurons in trigeminal nucleus caudalis and periaqueductal gray.
