超剂量荆防败毒散对小鼠肠道菌群及免疫相关指标影响的观察 被引量:2

摘要 目的:探讨超剂量荆防败毒散对小鼠肠道菌群及免疫相关指标的影响。方法:选用60只KM雌性小鼠,随机分成4组,每组15只:正常组(灌胃给予生理盐水)、一般剂量组(16.2 g·kg^(-1)荆防败毒散)、超剂量1组(48.6 g·kg^(-1)荆防败毒散)、超剂量2组(81 g·kg^(-1)荆防败毒散)。实验周期为15 d。观察小鼠体质量情况,给药15 d后,检测各组小鼠淋巴细胞数、胸腺指数、脾指数、肠道菌群及细菌移位现象等指标。结果:超剂量2组第14天体质量明显轻于正常组,球杆菌比为0.389;与正常组比较,超剂量2组小鼠肠道益生菌,相关免疫指标(淋巴细胞数、胸腺指数及脾指数)明显减少,且出现了3例细菌移位现象,均为移至肠系淋巴结。一般剂量组和超剂量1组小鼠体质量、免疫指标、肠道菌群及细菌移位情况均与正常组基本一致。结论:5倍一般剂量的超剂量荆防败毒散可导致小鼠免疫功能降低,肠道菌群失调。 OBJECTIVE To study effects of overdose Jingfang Baidu San on balance of mouse intestinal flora and immune functions of mice.METHODS Sixty Kunming mice were randomly divided into 4 groups:health control group(normal saline),normal dose group(16.2 g·kg^-1 Jingfang Baidu San),overdose group 1(48.6 g·kg^-1 Jingfang Baidu San),overdose group 2(81 g·kg^-1 Jingfang Baidu San),15 mice in each group.Mouse weight was observed at day1,5,10 and15.Lymphocyte count,gastric lavage,bacterial translocation and intestinal flora of mice were detected.RESULTS In overdose groups,body weights of mice were obviously less than in normal group 14 days later,and coccobacillary ratio was 0.389.Compared to normal group,overdose groups had significantly decreased intestinal probiotics and relevant immune parameters(lymphocyte count,thymus index and spleen index),and 3 mice of bacterial translocation all to mesenteric lymph nodes.Control group,normal dose group and overdose group 1 had basically consistent body weights,immune parameters,intestinal flora and bacterial translocation.CONCLUSION Administration of 81 g·kg^-1 Jingfang Baidu San can induced decreased immune functions and imbalance of intestinal flora.
