一种三苯甲烷类染料脱色菌液的制备及其脱色性能 被引量:4

Preparation and Decoloring Performance of a Triphenylmethane Dyes Decoloring Bacterial Liquid

摘要 通过加入适量三苯甲烷类染料驯化自然腐败发酵的肉汤培养基,分离出了一株耐氧厌氧脱色菌和一株兼性厌氧不脱色菌,根据其脱色菌株的培养和形态特征,初步确定该耐氧厌氧菌株为梭菌属的荚膜梭菌。驯化脱色菌液厌氧环境的形成和染料的毒性使该兼性厌氧菌处于劣势,而使耐氧厌氧脱色菌成为优势菌。兼性厌氧菌的存在不影响脱色菌的脱色性能,且能消耗环境中的溶解氧为厌氧菌提供有利的生长环境。通过人工接种驯化脱色菌液发酵肉汤培养基制成脱色菌液,分析了不同孔雀石绿和结晶紫含量的两组梯度脱色菌液脱色过程的紫外-可见光谱图。结果显示,孔雀石绿和结晶紫发色基团的结构被菌体破坏;脱色菌液在35℃条件下,24 h对孔雀石绿最大处理浓度约为168 mg/L,48 h对结晶紫最大处理浓度约为12 mg/L。 Through the acclimation of broth culture medium after natural putrefaction and fermentation by addition of a proper amount of malachite green and crystal violet, a strain of aerotolerant anaerobic decoloring bacteria and a strain of facuhative anaerobic bacteria were isolated. According to the cultural and morphologic characteristics of the decoloring bacterial strain, the aerotolerant anaerobic bacterial strain was preliminarily determined as clostridium capsule. The formation of anaerobic environment and the toxicity of dyes in the decoloring bacterial liquid after acclimation put the facultative anaerobic bacterial strain at a disadvantage and the aerotolerant anaerobic bacterial strain at an advantage. The facultative anaerobic bacterial strain did not affect the decoloring performance of decoloring bacteria, the consumption of dissolved oxygen by the facultative anaerobic bacteria provided a favourable environment for the growth of anaerobic bacteria. The decoloring bacterial liquid was prepared by artificial inoculation of the broth culture medium fermented with the decoloring bacterial liquid after acclimation, and the UV-Vis spectra of the two groups of decoloring bacterial liquids with different contents of malachite green and crystal violet during the decolorization processes were analyzed. The results showed that the decoloring bacteria destroyed the chromophore structure of malachite green and crystal violet, and the maximum treatment concentrations of malachite greenand crystal violet by the decoloring bacterial liquid at 35 ℃ were 168 mg/L at 24 h and 12 mg/L at 48 h respectively.
