摘要
目的探讨维生素E对甲基磺酸甲酯(MMS)诱导的大鼠精子畸形的影响。方法 12只雄性SD大鼠随机均分为模型组(MMS 40mg/kg灌胃5d后继予PBS灌胃43d)、治疗组(MMS40mg/kg灌胃5d后继予维生素E 150mg/kg灌胃43d)和对照组(予PBS灌胃48d)。采用心脏取血法处死大鼠,计数附睾精子数目,计算精子活率及精子畸形率,放免法检测血清睾酮水平,HE染色法观察睾丸结构,RT-PCR法检测生殖细胞Vasa、精母细胞Dmc1、支持细胞Sox9及血睾屏障Vim基因的表达。结果与对照组相比,模型组大鼠精子数量、活率、血清睾酮水平和生殖细胞Vasa基因表达降低,精子畸形率升高(P<0.05),生精小管出现损伤;与模型组相比,治疗组大鼠精子数量、活率、血清睾酮水平和生殖细胞Vasa基因表达升高,精子畸形率降低(P<0.05),生精小管损伤减轻。三组精母细胞Dmc1、支持细胞Sox9和血睾屏障Vim基因表达无统计学差异(P>0.05)。结论 MMS可能通过影响生殖细胞而导致精子畸形,维生素E有一定的治疗作用。
Objective To investigate the effects of vitamin E on methyl methanesulphonate(MMS)-induced teratozoospermia in rats.Methods Twelve male SD rats were equally randomized into 3groups of A(treated with MMS 40mg/kg for 5days followed by PBS gavage for 43days),B(treated with MMS 40mg/kg for 5days followed by gavage vitamin E 150mg/kg for 43days)and C(treated with PBS by gavage for 48 days as the control).Then the rats were sacrificed for counting the number of sperm and sperm motility and abnormality in epididymis.Serum level of testosterone was detected by radioimmunological method.The structure of the testis was observed by HE staining.The gene expressions of Vasa,Dmc1,Sox9 and Vim gene were detected by RT-PCR.Results Compared with group C,the number of sperm,sperm motility,serum level of testosterone,Vasa expression in germ cells were decreased(P〈0.05),sperm abnormality was increased(P〈0.05),and seminiferous tubule injury occurred in group A.Compared with group A,the number of sperm,sperm motility,serum level of testosterone,Vasa expression in germ cells were increased(P〈0.05),sperm abnormality was decreased(P〈0.05),and the injury of seminiferous tubule was alleviated in group B.There were no significant differences in the gene expressions of Dmc1,Sox9 and Vim among 3groups(P〉0.05).Conclusion MMS may cause sperm abnormalities through the injury of germ cells,which can be treated with vitamin E.
