摘要
目的研究辛伐他汀对慢性阻塞性肺病大鼠基质金属蛋白酶-9(MMP-9)及其抑制剂(TIMP-1)表达的影响。方法选用30只健康的雄性SD大鼠作为实验材料,将30只大鼠分为三组:A组10只,B组10只,C组10只。B组和C组构建慢阻肺模型,C组大鼠采用辛伐他汀治疗。比较各组大鼠的一般症状、肺功能、支气管肺泡灌洗液(BALF)细胞计数和血清MMP-9、TIMP-1水平。结果 4w后,各组大鼠的体质量、FEV0.3s、FVC、FEV0.3s/FVC、PEV和BALF细胞计数具有统计学差异(P<0.05)。A组大鼠MMP-9为(87.14±21.76)ng/ml,TIMP-1为(82.16±14.89)ng/ml,MMP-9/TIMP-1为(1.07±0.09);B组大鼠MMP-9为(241.58±38.94)ng/ml,TIMP-1为(167.92±19.76)ng/ml,MMP-9/TIMP-1为(1.44±0.21);C组大鼠MMP-9为(134.25±29.15)ng/ml,TIMP-1为(119.41±15.62)ng/ml,MMP-9/TIMP-1为(1.12±0.1),差异具有统计学意义(P<0.05)。结论辛伐他汀可以降低慢阻肺大鼠基质金属蛋白酶-9及其抑制剂的表达水平,并改善两者的比例失衡。
Objective To study the influence of simvastatin on matrix metalloproteinase-9 ( MMP-9 ) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in rats with chronic obstructive pulmonary disease. Methods 30 male SD rats were chosen as study objects, and all rats were divided into 3 groups: 10 in the group A ( control group) , 10 in the group B ( model group) and 10 in the group C ( simvastatin) . After 4 weeks, the symptoms, pul-monary function, cell counting of BALF and level of MMP-9 and TIMP-1 were compared among the 3 groups. Results After 4 weeks, the body mass, FEV0. 3s , FVC, FEV0. 3s/FVC, PEV and cell counting of BALF had statistic differ-ence among the 3 groups (P〈0. 05). In the group A, MMP-9 was (87. 14 ± 21. 76) ng/ml, TIMP-1was (82. 16 ± 14. 89) ng/ml, and MMP-9/TIMP-1 was (1. 07 ± 0. 09). In the group B, MMP-9 was (241. 58 ± 38. 94) ng/ml, TIMP-1 was ( 167. 92 ± 19. 76 ) ng/ml, and MMP-9/TIMP-1 was ( 1. 44 ± 0. 21 ) . In the group C, MMP-9 was (134. 25 ± 29. 15) ng/ml, TIMP-1 was (119. 41 ± 15. 62) ng/ml, and MMP-9/TIMP-1 was (1. 12 ± 0. 1). The difference had statistical significance (P〈0. 05). Conclusion Simvastatin can reduce the expression level of MMP-9 and TIMP-1 in rats with COPD, as well as regulate their ratio.
