摘要 【目的】探索靶向β_2-肾上腺素受体诱导骨肉瘤细胞凋亡的作用及相关分子机制。【方法】利用MTT法检测靶向β_2-肾上腺素受体对骨肉瘤细胞的生长抑制作用,流式细胞仪检测细胞周期阻滞及凋亡,Western blot法检测β_2-肾上腺素受体抑制剂ICI118,551处理后相关信号通路的变化。【结果】β_2-肾上腺素受体抑制剂对骨肉瘤细胞的生长抑制作用,流式细胞仪检测结果证明ICI118,551可显著诱导骨肉瘤细胞周期阻滞及凋亡,以p-ERK蛋白水平表达变化最为显著。【结论】靶向β_2-肾上腺素受体能显著诱导骨肉瘤细胞凋亡,其中ERK通路在靶向β_2-肾上腺素受体诱导的骨肉瘤细胞凋亡中具有重要作用。 [ Objective ] To study the anti-tumor effect by targeting 152-adrenoceptor in osteosarcoma and the underlying molecular mechanism. [ Methods ] MTF assay was used to detect the anti-proliferation effect in osteosarcoma following the cells were treated with a β2-adrenoceptor antagonist ICI118,551.Then, flow cytometry was employed to detect the cell cycle arrest and apoptosis. Finally, the concerning signal pathway wastested by Western blot. [ Result ] Targeting 132-adrenoceptor could inhibit osteosareoma cell proliferation. Significant G0/G1 cell cycle arrest and apoptosis were detected through flow cytometry following ICI118,551 incubation. Furthermore, the protein level of p-ERK was found down-regulated remarkably. [ Conclusion ] Together, targeting β2-adrenoceptor possesses potent anti-tumor effect in osteosarcoma cells. The ERK signaling pathway probably play a critical role in this process.
