摘要
从海参肠道中筛选出一株红酵母菌HS-J4,对其进行形态、培养特性分析及26SrDNA序列测定,建立了系统发育树,初步鉴定该菌株为胶红酵母。通过测定红酵母的生长曲线,确定红酵母的最佳发酵时间为24h,并检测了胶红酵母HS-J4对实验室配制的人工污水及海参养殖污水中亚硝酸盐和氨氮的清除能力。结果表明,HS-J4菌体对人工污水中高浓度的氨氮和亚硝酸盐氮均有良好的去除效果,清除率分别为42.3%和34.8%。应用于海参养殖污水时,HS-J4对水体中亚硝酸盐的去除率达到56.8%。筛选得到的胶红酵母HS-J4具有较高的硝化能力,可为养殖水质改良的实际生产提供有效参考。
A marine Rhodotorula strain named HS-J4 was isolated from sea cucumber intestine.It was identified as a Rhodotorula mucilaginosa strain analysed by the characterization of morphology,26 S rDNA and the phylogenic tree.The best fermentation time for cell growth of HS-J4 was 24h,and the scavenging activities for the nitrite and ammonia nitrogen in artificial wastewater and aquaculture wastewater was determined.The results showed that the removal rates of the ammonia nitrogen and nitrate could reach to 42.3%and 34.8%in the artificial wastewater,while the removal rate of nitrate was 56.8% in aquaculture wastewater for sea cucumber.The results showed that HS-J4 had high capability of removing the nitrite and ammonia nitrogen,which laid a good foundation for the further application in the aquaculture water.
