摘要
目的:探讨κ^-卡拉胶十一糖对小鼠单核/巨噬细胞RAW264.7细胞增殖活性的影响。方法:使用不同浓度的κ^-卡拉胶十一糖作用RAW264.7一段时间后,采用MTT比色法及检测LDH总量,测定细胞增殖率;通过测定LDH释放量及ATP水平,研究κ^-卡拉胶十一糖对细胞膜完整性及细胞活性的影响;绘制细胞生长曲线,观察κ^-卡拉胶十一糖对RAW264.7增殖能力的影响。结果:实验结果显示,使用100μg/m L的卡拉胶十一糖作用RAW264.7细胞24h后,细胞增殖率达到141.5%,且在此条件下,细胞膜完整性、细胞活性及细胞增殖能力的提高最为显著。但当作用时间延长为60h时,κ^-卡拉胶十一糖对细胞数量、细胞膜完整性、细胞活性及细胞增殖能力有影响。结论:在体外条件下,κ^-卡拉胶十一糖对小鼠单核/巨噬细胞RAW264.7的生长具有促进作用。
Objective:To investigate the effect of κ^-carrageenan oligosaccharides on cell proliferation in cultured mouse monocyte/macrophage cells RAW264.7. Methods:After the cells were treated with κ^-carrageenan oligosaccharides in different concentration at different time,the influence of κ^-carrageenan oligosaccharides on RAW264.7 proliferation rate was investigated by MTT assay and measurement of total LDH;the release quantity of LDH and ATP level was measured to investigate the effect of κ^-carrageenan oligosaccharides on cell membrane integrity and cell activity;cell growth curve was drew to investigate the effect of κ^-carrageenan oligosaccharides on cell proliferation in RAW264.7. Result:When the cells were stimulated 100μg/m L κ^-carrageenan oligosaccharides in 24 h,cell proliferation rates is 141.5%. Under this condition,the integrity of cell membrane,cell viability and cell proliferative potential is enhanced too. But when time of length was extended to 60 h,κ^-carrageenan oligosaccharides has the opposite effect for RAW264.7. Conclusion:κ^-carrageenan oligosaccharides can enhance the growth of mousemonocyte/macrophage cells RAW264.7.
