摘要
目的探讨T细胞免疫球蛋白域黏蛋白域蛋白-1(Tim-1)在儿童过敏性鼻炎(allergic rhinitis,AR)中的作用。方法选择42例AR患儿及同期30例健康儿童作为对照组。Real time qPCR检测AR患儿和对照组外周血单个核细胞(PBMC)Tim-1mRNA表达水平,流式细胞术检测Th2细胞比例,同时测定两组血清IgE浓度和外周血嗜酸性粒细胞比值。结果儿童AR患者Tim-1mRNA水平为0.76±0.16,明显高于对照组0.27±0.08;Th2细胞表达率(20.4%±7.6%),血清IgE浓度(255.78±68.29IU/m1)明显高于对照组相应检测值(12.3%±6.9%,16.22±6.01IU/m1),各项检测指标两组之间比较差异均有统计学意义(P〈0.05);且Tim-1mRNA水平与Th2细胞比例及血清IgE浓度呈显著正相关,而与外周血嗜酸性粒细胞比值间的相关性不明显。结论AR患儿Tirn-1mRNA高表达,并与Th2及临床指标IgE浓度密切相关,提示Tim-1可能通过调节Th2细胞分化参与儿童AR的发病过程,这为儿童AR临床治疗提供新的方向。
Objective To investigate the mechanism of T cell immunoglobulin domain mucin domain protein -1 (Tim-1) in children with allergic rhinitis (AR). Methods 42 children with AR and 30 healthy children were selected. Tim-1 mRNA levels in peripheral blood mononuclear cells (PBMC) were detected by real time qPCR,and Th2 cells were determined by flow cytometry. Serum IgE level and peripheral blood eosinophil ratio were also recorded. Results Tim-1 mRNA in children with AR were significantly increased (0.76±0.16) when compared to compared to control group (0.27±0.08) ,Th2 cells and serum IgE concentrations were significantly increased (20.4% ± 7.6 %, 255.78 ± 68.29 IU/ml) and Tim-1 mRNA levels Were positively correlated with Th2 cell and serum IgE concentrations,and no relationship with peripheral eosinophil ratio was observed. Conclusion Tim-1 mRNA was high in AR children and was closely related to IgE and Th2 cells. Thus,Tiro- l may be involved in the pathogenesis of AR children by regulating Th2 cells,providing a new direction for the clinical treatment of AR children.
