摘要
臭参地上部分富含蛋白质、黄酮及多糖,氨基酸态氮为0.820 g/100 g,总黄酮为7.980 mg/g,多糖为0.024 mg/g。以氨基酸态氮为评价指标,研究了臭参地上部分酶水解的优化条件,并考察了总黄酮、多糖含量的变化。结果表明水解最佳条件是:木瓜蛋白酶(≥800 U/mg)1.4 g/100 m L、时间8 h、p H7.5、温度60℃,水解后氨基酸态氮含量为1.271 g/100 g,总黄酮为3.832 mg/g,多糖为0.040 mg/g;菠萝蛋白酶(300 U/mg)≥5.6 g/100 m L、时间5 h、p H7.5、温度45℃,水解后氨基酸态氮含量为5.043 g/100 g,总黄酮为4.484 mg/g,多糖为0.036 mg/g。菠萝蛋白酶的水解效率高于木瓜蛋白酶。
The aerial parts of Codonopsis bulleyana Forrest was rich in protein,flavonoids and polysaccharide.The amino acid nitrogen was 0.820 g/100 g,flavonoids was 7.980 mg/g,polysaccharide was 0.024 mg/g. The optimum condition of enzyme hydrolysis for aerial part of Codonopsis bulleyana Forrest had been studied,amino acid nitrogen was used to as evaluation indexes. The content changes of flavonoids and polysaccharides have been investigated. The results showed that the optimum conditions of enzyme hydrolysis were obtained as follows:the papain(≥800 U/mg)content 1.4 g/100 m L,hydrolysis time 8 h,p H7.5,temperature 60 ℃,and after hydrolysis,the amino acid nitrogen content was 1.271 g/100 g,total flavonoids was 3.832 mg/g,polysaccharide was 0.040 mg/g.But the bromelain(300 U/mg)content ≥5.6 g/100 m L,hydrolysis time 5 h,p H7.5,temperature 45 ℃,and after hydrolysis,the amino acid nitrogen content was 5.043 g/100 g,total flavonoids was 4.484 mg/g,polysaccharide was 0.036 mg/g.In addition,The results showed that the papain higher than the bromelain on the hydrolysis efficiency.
