HIV/HCV合并感染者外周血中IP-10水平及其与肝损害的相关性 被引量:5

Relationship between the expression level of interferon-γ-inducible protein-10 and liver injury in HIV/HCV co-infected patients

摘要 目的研究炎性因子IP-10在人类免疫缺陷病毒/丙型肝炎病毒(HIV/HCV)合并者外周血中的表达水平及其对肝功能、HIV和HCV复制水平的影响。方法以未经抗病毒治疗的慢性丙型肝炎患者(HCV单一感染)、AIDS患者(HIV单一感染者,CD4^+T<200个/mm^3)和HIV/HCV合并感染者(CD4^+T<200个/mm3)为研究对象,并以健康志愿者作对照,抽取外周静脉血,分离血浆,ELISA检测血浆中IP-10的表达量,real-time PCR定量检测血浆中HIV病毒载量、HCV病毒载量,同时检测丙氨酸氨基转移酶(ALT)水平。结果相对于健康对照组,HIV单一感染组和HCV单一感染组的血浆IP-10水平显著升高(均为P<0.01);HIV/HCV合并感染组IP-10表达水平高于HCV单一感染组(P<0.01),HIV单一感染组稍高于HCV单一感染组(P<0.01);IP-10与ALT、HIV病毒载量及HCV病毒载量间无明显的相关性(均为P>0.05)。结论 HIV/HCV合并感染者外周血中高表达炎性因子IP-10,但IP-10对病毒复制及肝脏炎症并无明显影响。 Objective To study the difference of IP-10 expression level in human immunodeficiency virus/hepatitis C virus(HIV/HCV) coinfected patients and in mono-infected patients, the relationship between IP-10 expression level and liver injury, and the impact of IP-10 on HIV and HCV replication. Methods The treatment naive individuals including chronic hepatitis C patients(HCV mono-infected patients, 41 cases), HIV mono-infected patients(CD4~+T200 cells/mm^3, 45cases)and HIV/HCV-coinfected patients(CD4~+T 200 cells/mm^3, 41 cases) were recruited for this study, and 23 healthy volunteers were enrolled in control group. Venous blood was collected from individuals and then plasma were isolated, and IP-10 level was determined by ELISA. ALT and HCV RNA viral load were tested in HCV mono-infected patients and HIV /HCV-coinfected patients. HIV RNA viral load was tested in HIV mono-infected patients and HIV/HCV-conifected patients.Results Comparing with healthy control group,IP-10 level was significantly higher in HIV mono-infected group and HCV mono-infected group(all P〈0.01). IP-10 level of HIV/HCV-coinfected group was higher than that of HCV mono-infected group( P〈0. 01). There was no correlation between IP-10 level and ALT level(P〈0.05). The level of IP-10 showed no correlation with HIV-RNA viral load and HCV-RNA viral load(all P〈0.05). Conclusion IP-10 expression level in the peripheral blood of HIV/HCV-coinfected patients was high, but it showed no impact on the virus replication and liver inflammation.
