苏南地区汉族人群不稳定型心绞痛患者IL-10基因启动子819C/T多态性分析

Polymorphism analysis of interleukin-10 gene promoter 819C/T polymorphism in patients with unstable angina pectoris among Han population of Sunan region

目的探讨苏南地区汉族人群白介素-10(IL-10)基因启动子819C/T多态性与不稳定型心绞痛(UAP)发病的相关性。方法选取2010年6月至2013年2月在溧阳市人民医院心内科及武进人民医院心内科住院的UAP患者146例作为A组,另选取同期行冠状动脉造影排除冠心病患者153例作为B组。采用DNA试剂盒提取两组患者外周静脉血DNA,用回收试剂盒回收PCR产物,进一步测序鉴定IL-10基因819C/T多态性,检测血糖和血脂等生化指标。结果IL-10基因启动子819C/T在A、B组中均存在CC、CT和TT 3种基因型,且基因分型符合Hardy-Weinbery平衡;A组中3种基因型分布频率及等位基因分布频率与对照组比较,差异均无统计学意义(P>0.05);调整对多个危险因素行多元logistic回归性分析,提示IL-10基因启动子819C/T 3种基因型与UAP的发病仍无相关性。结论 IL-10基因819C/T多态性与苏南地区汉族人群UAP的发病可能无相关性。

Objective To investigate the relationship between interleukin-10（IL-10）gene promoter 819C/T polymorphism and unstable angina pectoris（UAP） in the Chinese Han population of Sunan region. Methods From June 2010 to February 2013,a total of 146 patients with UAP（group A） and other 153 healthy subjects without coronary artery disease by coronary arterial angiography（group B） were selected into the study. The DNA kit was adopted to extract DNA from peripheral venous blood,and the PCR products were recovered by the recycle reagent kit. The IL-10 gene polymorphisms were identified by further sequencing.The biochemical indicators of blood sugar and blood fat were detected. Results The three genotypes CC,CT and TT in IL-10 gene promoter 819C/T existed in the group A and B,moreover the genotyping conformed to the Hardy-Weinbery balance;the 3kinds of genotype distribution frequency and allele distribution frequency in the group A had no statistical difference compared with the control group（P〉0.05）;the multivariate Logistic regression analysis by adjusting multiple risk factors indicated that there was no significant correlation between three genotypes of IL-10 gene promoter 819C/T and UAP pathogensis. Conclusion The IL-10 gene 819C/T polymorphism may have no correlation with UAP pathogensis among the Chinese Han population of Sunan region.
