摘要
目的:研究α-硫辛酸对缺血再灌注脑组织神经功能的保护效应。方法:选择72只雄性SD大鼠作为实验动物,随机分为对照组、模型组、干预组,采用线栓法建立脑组织缺血再灌注模型,给予α-硫辛酸进行干预。干预后6h、12h、24h、48h时,取脑组织测定氧化应激反应程度、NF-kB表达量以及NLRP3炎性小体的功能。结果:模型组大鼠脑组织中MDA、NF-kB、NLRP3、ASC、Caspase-1、IL-1β、IL-18的含量显著高于对照组,SOD、GSH-Px的含量显著低于对照组;干预组大鼠脑组织中MDA、NFkB、NLRP3、ASC、Caspase-1、IL-1β、IL-18的含量显著低于模型组,SOD、GSH-Px的含量显著高于模型组。结论:α-硫辛酸能够通过减轻氧化应激反应程度、抑制NLRP3炎性小体功能的途径来保护缺血再灌注脑组织的神经功能。
Objective:To study the protective effect ofα-lipoic acid on neurological function of ischemia-reperfusion brain tissue.Methods:A total of 72 male SD rats were selected as experimental animals and randomly divided into control group,model group and intervention group,suture-occluded method was used to establish cerebral ischemia-reperfusion models,andα-lipoic acid was used for intervention.6h,12 h,24hand 48 hafter intervention,brain tissue was collected to detect the degree of oxidative stress response,NF-kB expression level and NLRP3 inflammasome function.Results:MDA,NF-kB,NLRP3,ASC,Caspase-1,IL-1βand IL-18 content in brain tissue of model group were significantly higher than those of control group while SOD and GSH-Px content were significantly lower than those of control group;MDA,NF-kB,NLRP3,ASC,Caspase-1,IL-1βand IL-18 content in brain tissue of intervention group were significantly lower than those of model group while SOD and GSH-Px content were significantly higher than those of model group.Conclusion:Sα-lipoic acid can reduce the degree of oxidative stress response and inhibit NLRP3 inflammasome function to protect the neurological function of ischemiareperfusion brain tissue.
