摘要
目的:观察电针干预对束缚应激模型大鼠行为学及血清中白细胞介素6(IL-6)与可溶性白细胞介素6受体(s IL-6R)表达的影响,探讨其相关免疫学机制。方法:将60只SD大鼠随机分为对照组、模型组与电针组,每组20只。对照组群养;模型组采用孤养结合慢性束缚应激方式,每日束缚6 h(9:00—15:00);电针组在每日束缚前给予电针刺激"百会""印堂";束缚期间3组均禁食禁水,其余时间自由饮食饮水。采用旷场和体质量实验进行行为学评价,并在第7天、28天、35天采用Bio-Plex检测大鼠血清中IL-6、ELISA检测SIL-6R水平。结果:1行为学:与对照组相比,模型组大鼠体质量及活动性均显著降低(P<0.01),电针组大鼠体质量及活动性表达无显著差异(P>0.05)。2血清IL-6表达:实验day7,与对照组相比,模型组IL-6表达显著增加(P<0.01),电针组IL-6表达无显著差异(P>0.05);与模型组相比,电针组IL-6表达显著降低(P<0.05)。实验day28,与对照组相比,模型组IL-6表达显著增加(P<0.01),电针组IL-6表达无显著差异(P>0.05);与模型组相比,电针组IL-6表达显著降低(P<0.01)。实验day35,3组大鼠之间IL-6表达无显著差异(P>0.05)。3血清s IL-6R表达:实验day7,3组大鼠之间s IL-6R表达无显著差异(P>0.05);实验day28,与对照组相比,模型组s IL-6R表达显著增加(P<0.01),电针组s IL-6R表达无显著差异(P>0.05);与模型组相比,电针组s IL-6R表达显著降低(P<0.05)。实验day35,3组大鼠之间s IL-6R表达无显著差异(P>0.05)。结论:慢性束缚应激模型大鼠血清IL-6、s IL-6R表达显著增加,并随时间出现相应变化,电针干预可对其进行显著调控,与行为学症状改善一致,这可能是电针抗抑郁的作用机制之一。
Objective: To explore the immune mechanism on behavior of rats with restraint stress and the expression of interleukin 6( IL- 6) and soluble interleukin 6 receptor( s IL- 6R) in serum by electroacupuncture. Methods: Sixty SD rats were equally divided into a control group,a model group and an acupuncture group. The control group was bred in group; in the model group the rats were with the condition of chronic restraint stress and separation,daily bound for 6h( 9: 00- 15: 00); electroacupuncture group was stimulated at"Baihui"and"Yintang"before daily bound; solids and liquids were inhibited during the period of bound,but free for the rest of time. The behavior was evaluated by spacious field and body weight,and the serum IL- 6and s IL- 6R levels were detected by Bio- Plex and ELISA respectively on the 7th,28 th,35th day. Results:Effects on Behavior: compared with the control group,the body weight and activity were significantly decreased in model group( P〈0. 01),and it showed no difference in EA group( P〈0. 05). Effects on IL- 6: After 7days,compared with the control group,the model group was significantly increased( P〈0. 01) and there was no difference in EA group; compared with the model group,EA group was significantly decreased( P〈0. 05); after28 days,compared with the control group,model group was significantly increased( P〈0. 01) and there was no difference in EA group; compared with the model group,IL- 6 in EA group was significantly decreased( P〈0. 01); after 35 days,there was no significant difference among three groups( P〈0. 05). Effects on s IL- 6R in serum: After 7 days,there was no significant difference among three groups( P〈0. 05). After 28 days,compared with the control group,model group was significantly increased( P〈0. 01) and there was no difference in EA group; compared with the model group,s IL- 6R in EA group was significantly decreased( P〈0. 05); there was no difference among three groups on the 7th and 35 th day( P〈0. 05). Conclusion: The expressions of IL- 6and s IL- 6R are significantly increased over time in serum of rats with chronic restraint stress. EA intervention can significantly regulate them,consisting with the improvement of behavior,which may be one of the mechanisms of anti- depression by electroacupuncture.
