摘要
对新西兰青霉(Penicillium novae-zeelandiae)红色素的发酵条件进行优化,并对其组分进行初步分析.研究结果表明,新西兰青霉红色素的最佳发酵条件是:用1mL的接种量(孢子悬液浓度为2×10~7 mL^(-1))接种于300mL的初始pH为6.0的PD培养基内(葡萄糖质量浓度为18g·L^(-1)),30℃150r·min^(-1)振荡培养40h,之后将其静置于30℃的培养箱内进行培养.经过优化后,红色素的最高产量达到2.834 4g·L^(-1).高效液相色谱和薄层层析分析表明,在260nm的检测波长下,该红色素分别在16.667min,18.120min,19.895min,21.022min处出现4个显著的吸收峰.
In this paper,the fermentation process was designed according to the yield of red pigments produced by Penicilliumnovae-zeelandiae HSD07 B.The components of red pigment were also analyzed.The results showed that the optimal parameters are:PD medium,500 mL flask containing 300 mL medium,the inoculum volume 1 mL(Spore concentration 2×10^7 mL^(-1)),initial pH 6.0,rotary speed of 150r·min^(-1),temperature 30 ℃,oscillating time 40 h.Under the optimal conditions,the maximum concentration of red pigment is 2.283 44g·L^(-1).All components of red pigments were analyzed by high performance liquid chromatography(HPLC)and thin-layer chromatography(TLC),and the retention time of four major components were 16.667 min,18.120 min,19.895 min,21.022 min,respectively.
